当归挥发油对自发性高血压大鼠NO、vWF、EMPs表达水平影响

摘    要:目的 通过观察当归挥发油对自发性高血压大鼠(Spontaneously hypertensive rats,SHR)血管内皮活性物质表达的影响来探讨其降压的作用机制。方法 将8周龄雄性Wistar大鼠作为正常组,每组6只。并将同周龄自发性高血压大鼠(SHR)随机分组,各组分别为模型组、缬沙坦组、当归低、中、高剂量组,每组6只。模型组和正常组给予等量蒸馏水,连续灌胃4周。采用无创血压检测系统测定给药前后大鼠尾动脉收缩压;给药4周后麻醉大鼠,腹主动脉采血,酶联免疫吸附测定(Enzyme-linked immunosorbent assy,ELISA)检测大鼠血清中一氧化氮(Nitric oxide,NO)、血管假血友病因子(von Wilebrand factor,vWF)、内皮细胞膜微粒(Endothelial Microparticles,EMPs)表达水平。结果 ELISA结果显示:与正常组比较,模型组EMPs、vWF水平明显升高(P < 0.01),NO水平明显降低(P < 0.01),差异具有显著性;与模型组比较,各给药组EMPs、vWF水平明显降低(P < 0.01),NO水平明显升高(P < 0.01),差异具有显著性。结论 当归挥发油具有显著的降压效果,其降压作用可能与抑制血清中的EMPs、vWF以及促进了血清中的NO水平有关。

Abstract:Objective The mechanism of antihypertensive effect was investigated by observing the effects of volatile oil of Angelica sinensis on the expression of vascular endothelial active substances in Spontaneously hypertensive rats (SHR).Methods Eight-week-old male Wistar rats were used as the normal group, with 6 rats in each group. Spontaneously hypertensive rats (SHR) of the same age were randomly divided into groups, including model group, valsartan group, and Angelica low, medium and high dose groups, with 6 rats in each group. The model group and the normal group were given an equal amount of distilled water and continuously administered for 4 weeks. The non-invasive blood pressure detection system was used to measure the systolic blood pressure of the rat tail artery before and after administration. After 4 weeks of administration, the rats were anesthetized, the blood was collected from the abdominal aorta, and the serum was detected by enzyme-linked immunosorbent assy (ELISA). Nitric oxide (NO), von Wilebrand factor (vWF), and Endothelial Microparticles (EMPs) expression levels.Results The results of ELISA showed that compared with the normal group, the levels of EMPs and vWF in the model group were significantly increased (P < 0.01), and the levels of NO were significantly decreased (P < 0.01). The difference was significant. Compared with the model group, EMPs in each drug group, the level of vWF was significantly decreased (P < 0.01), and the level of NO was significantly increased (P < 0.01). The difference was significant.Conclusion Angelica sinensis volatile oil has a significant antihypertensive effect, and its antihypertensive effect may be related to the inhibition of serum EMPs, vWF and the promotion of NO levels in serum.
