CBIQ及多巴酚丁胺对氧化损伤的肺泡上皮细胞活性的影响

摘    要:目的研究CBIQ及多巴酚丁胺对氧化损伤的肺泡上皮细胞株A549增殖活性的影响。方法应用四甲基偶氮唑蓝(MTT)法检测不同浓度CBIQ、不同浓度多巴酚丁胺及两种药物共同作用对过氧化氢(H2O2)损伤的肺泡上皮细胞株A549增殖活性的影响。结果 H2O2损伤后的A549细胞经不同浓度CBIQ及多巴酚丁胺处理后,细胞死亡数均明显减少。浓度为1×10-2,1×10-3mmol/L的CBIQ组平均细胞存活率分别为(76.12±0.81)%,(71.31±1.00)%,较损伤组(46.34±0.90)%明显增高(P<0.01);浓度为1×10-2,1×10-3,1×10-4mmol/L的多巴酚丁胺组平均细胞存活率分别为(70.53±0.22)%(、67.82±0.29)%(、63.53±0.61)%,较损伤组(32.50±0.12)%明显增高(P<0.01);但CBIQ及多巴酚丁胺共同干预组平均细胞存活率(57.31±0.60)%较多巴酚丁胺组(69.61±0.92)%及CBIQ组(63.20±0.80)%均明显减少,其中CBIQ联合多巴酚丁胺共同干预组与多巴酚丁胺组比较差异有统计学意义(P<0.05)。结论在一定浓度范围内,CBIQ及多巴酚丁胺对氧化损伤的A549细胞生长有浓度依赖性的保护作用,但两者共同作用后其保护作用减弱。

Effect of CBIQ and Dobutamine on oxidatively damaged human lung epithelial cell line A549
Abstract:Objective To explore the effect of 4-Chloro-benzo[F]isoquinoline(CBIQ) and/or Dobutamine on oxidatively damaged human lung epithelial cell line A549.Method After cultured with different doses of CBIQ and/or Dobutamine,the proliferative activity of the normal A549 cells and the oxidatively damaged cells were both determined by MTT assay.Results After A549 cells were treated with different concentrations of CBIQ or Dobutamine,dose-dependent protection was demonstrated.The cell viability rate of the group by 1×10-2 and 1×10-3 mmol/L CBIQ were respectively(76.12±0.81)% and(71.31±1.00)%,which were markedly higher than the rate of the injury group [(46.34±0.90)%,P<0.01].After they were treated with different concentrations of Dobutamine,the cell viability rate of the group by 1×10-2,1×10-3 and 1×10-4 mmol/L Dobutamine were respectively(70.53±0.22)%,(67.82±0.29)% and(63.53±0.61)%,which were also markedly higher than the rate of the injury group [(32.50±0.12)%,P<0.01].But the cell viability rate of the group treated with CBIQ and Dobutamine was(57.31±0.60)%,which was lower than that in group with only Dobutamine [(69.61±0.92)%,P<0.05]or CBIQ [(63.20±0.80)%,P<0.05].Conclusion In a certain concentration range,CBIQ and Dobutamine can dose-dependently protect human lung epithelial cells from oxidative damage.However,the protective effect of these two factors together was weakened.
