慢性疲劳大鼠模型的建立及其对肝功能的影响

摘    要:目的建立大鼠慢性疲劳模型,观察慢性疲劳对大鼠肝功能的影响。方法100只大鼠随机分为正常对照组和慢性疲劳模型组,两组又按强制游泳周数各分为5个亚组,慢性疲劳模型组大鼠每日强制游泳2h,慢性疲劳模型各亚组造模前及造模最后1d观测大鼠的力竭游泳时间,正常对照组大鼠除在疲劳模型组大鼠游泳期间给予禁食禁水外其他不作干预。实验结束后大鼠腹主动脉采血,检测肝功能相关指标:谷丙转氨酶(ALT)、谷草转氨酶(AST)、胆红素(TBIL)、总蛋白(TP)、白蛋白(ALB)、球蛋白(G)。结果慢性疲劳模型组大鼠造模2周以后其力竭游泳时间显著减少(P〈0.01),并随着造模时间延长至5周,大鼠力竭游泳时间有减少的趋势,但无统计学意义(P〉0.05)。与正常对照组比较,慢性疲劳模型组大鼠造模2周时血清AST出现显著升高(P〈0.05),并持续到5周(P〈0.01);大鼠造模3周时血清ALT出现显著性升高(P〈0.05),并持续到5周(P〈0.01);其他肝功能相关指标未见异常。结论大鼠强制游泳2周可造成慢性疲劳模型,并造成大鼠肝功能损伤。

Abstract:Objective To observe the influence of chronic fatigue on rat's liver function.Method 100 rat were randomly divided into two groups:the control group and the model group.Then each group was divided into 5 sub-groups according to the number of weeks of forced-swimming.The model group was given forced-swimming exercise 2 hours a day,7days a week.The exhausting swimming time was recorded before and after experiment.Also ALT,AST,TBIL,TP,ALB and G were examined at the end of every week.The control group was kept fasting and water privation during the model group was swimming.Result Compared with the control group,the exhausting swimming time of the model group decreased significantly(P < 0.01)since the 2nd week and had a decreasing tendency without significant difference(P > 0.05).There was a significant increase level of AST(P < 0.01)since the 2nd week in the fatigued rat compared with the control ones.The ALT level also indicated a significant increase from the 3rd week to the 5th week(P < 0.05).There was no significant change of levels of TBIL,TP,ALB and G(P > 0.05).Conclusion 2 weeks forced-swimming can establish the chronic fatigue model of rat and induce liver function injury.
