卵巢癌患者组织中miR-29b、miR-187表达与预后关系

摘    要:目的分析miR-29b和miR-187在卵巢癌患者组织中表达情况,探讨两者表达与卵巢癌患者病理特征和预后关系。方法选择2013年1月-2016年5月金华市中心医院收治的卵巢癌患者76例作为研究对象,另选择76例正常卵巢组织作为对照。采用实时荧光定量(qRT-PCR)法检测组织中miR-29b与miR-187表达水平,分析两者表达与卵巢癌患者病理特征和预后关系。结果卵巢癌组织中miR-29b表达水平显著低于正常卵巢组织(P<0.05),miR-187表达水平显著高于正常卵巢组织(P<0.05)。miR-29b和miR-187异常表达与卵巢癌患者年龄、病理分型等无关(均P>0.05),miR-29b低表达组低分化、FIGOⅢ期、有腹水以及淋巴结转移患者比例显著高于miR-29b高表达组(均P<0.05),miR-187高表达组低分化、FIGOⅢ期、有腹水以及淋巴结转移患者比例显著高于miR-187低表达组(均P<0.05)。卵巢癌组织中miR-29b和miR-187表达水平呈负相关(r=0.672,P<0.05)。miR-29b低表达组、miR-187高表达组患者3年总生存率分别显著低于miR-29b高表达组以及miR-187低表达组(P<0.05)。低分化、高FIGO分期、淋巴结转移、miR-29b低表达以及miR-187高表达是影响卵巢癌患者不良预后的独立危险因素(P<0.05)。结论miR-29b在卵巢癌组织中低表达,miR-187高表达,两者与卵巢癌分化程度、淋巴结转移等临床病理特征及预后有关,可能共同影响卵巢癌不良预后发生。

Abstract:Objective To analyze the expressions of miR-29b and miR-187 in ovarian cancer tissues,and to explore the relationships between the expressions of miR-29b and miR-187 and the pathological characteristics,prognosis of ovarian cancer patients.Methods 76 patients with ovarian cancer admitted to our hospital from January 2013 to May 2016 were selected as the study subjects,and 76 normal ovarian tissues were selected as the control group.Real-time fluorescence quantitative assay(qRT-PCR)was used to detect the expression levels of miR-29b and miR-187 in tissues,and the relationships between the expressions of miR-29b and miR-187 and the pathological characteristics,prognosis of ovarian cancer patients were analyzed.Results The expression of miR-29b in ovarian cancer was significantly lower than that in normal ovarian tissues(P<0.05),and the expression of miR-187 was significantly higher than that in normal ovarian tissues(P<0.05).The abnormal expressions of miR-29b and miR-187 were not related to the age and pathological type of ovarian cancer patients(P>0.05).However,the proportion of poor differentiation,FIGOⅢstage,with ascites and lymph node metastasis in miR-29 low expression group is higher than miR-29 high expression group(P<0.05).And the proportion of poor differentiation,FIGOⅢstage,with ascites and lymph node metastasis in miR-187 high expression group is higher than miR-187 low expression group(P<0.05).There was a negative correlation between the expressions of miR-29b and miR-187 in ovarian cancer(r=0.672,P<0.05).The 3-year overall survival rates of the miR-29b low-expression group and the miR-187 high-expression group were significantly lower than those of the miR-29b high-expression group and the miR-187 low-expression group(P<0.05).Poor differentiation,high FIGO stage,lymph node metastasis,low expression of miR-29b and high expression of miR-187 were independent risk factors for poor prognosis of ovarian cancer patients(P<0.05).Conclusion The expression of miR-29b is low and miR-187 is high in ovarian cancer.Both of them are related to clinicopathological characteristics and prognosis of ovarian cancer,such as differentiation degree,lymph node metastasis,and may affect the occurrence of poor prognosis of ovarian cancer.
