血管瘤患儿血清miR-126水平与瘤体增生的关系

摘    要:目的检测血管瘤患儿血清miR-126水平,分析其与血管瘤瘤体增生的关系。方法选取2016年2月-2019年4月厦门大学附属妇女儿童医院小儿外科收治的血管瘤患儿112例,分为增生期组(78例)和退化期组(34例)。另选取同期确诊为血管畸形的52例患儿为血管畸形组。应用HE染色观察血管组织病变情况,进行微血管密度(MVD)计数,应用qRTPCR法检测血清miR-126水平,应用酶联免疫吸附法(ELISA)检测血清VEGF、缺氧诱导因子-1(HIF-1)、血管生成素1(Ang-1)水平。结果CD34阳性表达主要位于血管瘤患儿血管内皮细胞胞质中。血管畸形组、退化期组、增生期组瘤体组织MVD依次升高,差异有统计学意义(P<0.05),组间两两比较差异均有统计学意义(均P<0.05)。3组血清miR-126水平比较差异有统计学意义(P<0.05)。血管畸形组和退化期组血清HIF-1α、VEGF、Ang-1水平比较,差异无统计学意义(P>0.05)。与退化期组、血管畸形组相比,增生期组血清Ang-1水平显著降低,差异有统计学意义(P<0.05),HIF-1α、VEGF水平显著升高,差异有统计学意义(P<0.05)。增生期血管瘤患者血清miR-126水平与MVD,血清VEGF、HIF-1α水平呈负相关(P<0.05),与血清Ang-1水平呈正相关(P<0.05)。结论miR-126在增生期血管瘤患者血清中低表达,可能与瘤体组织血管生成和增生过程有关。

Relationship between serum miR-126 level and neoplasia in children with hemangioma

Abstract:Objective To detect the level of serum miR-126 in children with hemangioma,analyze the relationship with neoplasia.Methods From February 2016 to April 2019,112 children with hemangioma treated by Department of Pediatric Surgery,Women and Children's Hospital Affiliated to Xiamen University were selected and divided into proliferative period group(78 cases)and degradation period group(34 cases);52 children diagnosed as vascular malformation during the same period were selected as vascular malformation group.HE staining was used to observe vascular tissue lesion,microvessel density was counted,qRT-PCR was used to detect serum miR-126 level,ELISA was used to detect the levels of serum VEGF,hypoxia-inducible factor(HIF-1),angiogenin 1(Ang-1).Results Positive expression of CD34 mainly located in cytoplasm of vascular endothelial cells in children with hemangioma.MVD values in vascular malformation group,degradation period group,and proliferative period group increased gradually(P<0.05),there was statistically significant difference between every two groups(P<0.05).There was statistically significant difference in serum miR-126 level among the three groups(P<0.05).There was no statistically significant difference in the levels of serum HIF-1α,VEGF,and Ang-1 between vascular malformation group and degradation period group(P>0.05).Compared with degradation period group and vascular malformation group,serum Ang-1 level in proliferative period group decreased significantly(P<0.05),the levels of HIF-1α and VEGF increased significantly(P<0.05).In proliferative period group,serum miR-126 level was negatively correlated with MVD,serum VEGF and HIF-1α levels(P<0.05),serum miR-126 level was positively correlated with serum Ang-1 level(P<0.05).Conclusion The expression of miR-126 is low during proliferative period in children with hemangioma,which may be related to angiogenesis and proliferation of tumor tissue.
