丹参单体、三氟拉嗪对视网膜色素上皮细胞增殖抑制的研究

摘    要:目的研究丹参单体(IH764-3)和三氟拉嗪对体外培养的兔视网膜色素上皮(retinal pigment epithelial,RPE)细胞增殖的抑制作用。方法将传代培养的RPE细胞分别加药培养24h、48h、72h、96h、120h,应用二甲基偶氮唑盐(MTT)比色法检测不同剂量丹参单体和三氟拉嗪对RPE细胞的抑制作用,进一步检测两药半数抑制剂量(IC50)联合应用对RPE细胞的抑制作用。结果丹参单体和三氟拉嗪对RPE细胞的增殖均有抑制作用,呈时间及剂量依赖性,各剂量及各时间点相比差异均有显著统计学意义(均为P<0.01),丹参单体的IC50为15.22μmol·L-1(2.4mg·L-1),三氟拉嗪的IC50为18.09μmol·L-1。联合用药120h对RPE的抑制率达87.23%,联合用药后各时间点与单独应用丹参单体及三氟拉嗪的抑制率比较,差异也均有显著统计学意义(均为P<0.01)。结论丹参单体和三氟拉嗪对RPE细胞的增殖均有抑制作用,两药联合后可产生协同作用,为以后两种药物的联合体内试验提供了一定的实验基础。
