舒洛地特对大鼠脑缺血再灌注损伤后脑组织的保护作用及机制

摘    要:目的探讨舒洛地特对大鼠脑缺血再灌注后脑组织的保护作用及机制。方法将72只Wistar大鼠随机分为3组即假手术组、模型组、舒洛地特干预组,采用改良Zea Longa[1]线栓法制备大鼠大脑中动脉局灶性脑缺血模型,应用免疫组化法检测肿瘤坏死因子-а(TNF-а)及C-反应蛋白(CRP)的表达。结果大鼠局灶性脑缺血再灌注后缺血脑组织中TNF-a及CRP含量增加(P<0.01)。与模型组相比较,舒洛地特组TNF-a及CRP的含量减少,脑组织肿胀减轻,死亡神经元明显减少,神经功能缺损评分降低(P<0.05)。结论舒洛地特对大鼠脑缺血再灌注损伤有一定的保护作用,其机制可能是多方面的。

Abstract:ObjectiveTo investigate the neuroprotective effects and mechanism of sulodexide on focal cerebral ischemia and ischemia-reperfusion injury in rats.MethodsSeventy-two Wistar rats were randomly divided into three groups,sham-operative group,model group and sulodexide group.The model of middle cerebral artery occlusion(MCAO) was made by the method of modified Zea Longa.The expressions of TNF-α and CRP were detected by immunohistochemical analysis.ResultsThe expressions of TNF-α and CRP in the ischemic territory increased after cerebral ischemia(P<0.01).Compared with the model group,the expression of TNF-α and CRP decreased in sulodexide group,the swelling of cerebral lightened,the number of death nerve cell decreased,and Longa’s score reduced(P<0.05).ConclusionSulodexide could protect the cerebral after focal cerebral ischemia,the mechanism is complicated.
