三磷酸腺苷生物荧光法和细菌培养法监测中度危险医疗用品安全性研究

摘    要:目的:采用三磷酸腺苷(ATP)生物荧光法和细菌培养法监测中度危险医疗用品,评估存放中的中度危险医疗用品的安全性,并进行效果分析。方法:采用ATP生物荧光法和细菌培养法对清洗消毒后的中度危险医疗用品进行采样和监测,将清洗消毒后的中度危险医疗用品分别采用医用塑料自封袋、棉布双层布袋和聚乙烯塑料袋3种包装,存放0 d、7 d、14 d、1个月、3个月、6个月、9个月和12个月后进行8个时间段的采样和监测,对比分析两种方法的监测效果。结果:细菌培养法的8个时间段检测中,医用塑料自封袋和聚乙烯塑料袋均未检出有害细菌;棉布双层布袋在存放3个月时检出干燥棒状杆菌1个,存放6个月时检出四联球菌2个;3种包装微生物监测合格率比较无差异。ATP检测医用塑料自封袋中的高效消毒物品ATP值1年内均为正常值;棉布双层布袋存放1个月时ATP值超出正常值;聚乙烯塑料袋0 d至1年的8个时间段ATP值均不合格,棉布双层布袋与聚乙烯塑料袋比较无差异;7 d,14 d、1个月、3个月、6个月、9个月和12个月医用塑料自封袋与棉布双层布袋和聚乙烯塑料袋比较,差异有统计学意义(F=3.142,F=3.900,F=9.556,F=7.705,F=10.596,F=10.282,F=14.204;P<0.05)。结论:ATP检测法结果检出及时,细菌培养法结果检出则需2 d;ATP检测值为升高或下降,细菌培养检测可检出具体细菌;ATP检测快速且有效,但无法检出具体致病菌。

Application of ATP bioluminescence assay and bacterial culture method in safety monitor of medical supplies with moderate risk

Abstract:Objective:To analyze the detection effects of adenosine triphosphate(ATP)bioluminescence assay and bacterial culture method so as to assess the safety of medical supplies with moderate risk in storage.Methods:Using the ATP bioluminescence assay and bacterial culture method to sample and monitor medical supplies with moderate risk after they were cleaned and sterilized.These medical supplies were packaged in three kinds of bags included medical plastics valve bag,double-layer cotton bag and polyethylene plastic bag.After they were stored,the sampling and monitoring were implemented at 8 time points included 0th d,7th d,14th d,1th m,3th m,6th m,9th m and 12th m.And the monitoring effects of the two methods were compared and analyzed.Results:The results of bacterial culture method for 8 time points indicated that the harmful bacteria were not detected in medical plastics valve bag and polyethylene plastic bag,and 1 drying Corynebacterium was found in double-layer cotton bag at 3th month and 2 Tetragenococcus were found in that at 6th month.The differences of qualified rates of microbial monitoring among three kinds of packages were no significant.The results of ATP fluorescence monitoring indicated that the ATP values of efficient disinfectant materials in medical plastics valve bagin 1 year were maintained within normal range,and the ATP value exceeded normal value when they were stored in double-layer cotton bag at 1th month,and all of ATP values of medical supplies in polyethylene plastic bagwere disqualification in 8 time points,from 0th d to 12th month,and there was no significant in ATP value between double-layer cotton bag and polyethylene plastic bag.The differences of ATP values among medical plastics valve bag,double-layer cotton bag and polyethylene plastic bag at 7th d,14th d,1th month,3th month,6th month and 9th month were significant(F=3.142,F=3.900,F=9.556,F=7.705,F=10.596,F=10.282,F=14.204,P<0.05).Conclusion:ATP detection method can obtain results in time,while bacterial culture method need 2d for obtaining result.The detection values of ATP detection method only appear increasing or decreasing,while bacterial culture method can detect specific amount of bacteria.ATP detection method is quick and effective,while it can’t detect specific pathogenic bacterium.
