不同何首乌特征图谱的初步研究及发酵何首乌主要成分的初步鉴定

摘    要:目的:通过对何首乌、制何首乌及发酵何首乌特征图谱的初步研究,比较何首乌、制何首乌及发酵何首乌化学成分的差异,并运用HPLC-TOF/MS技术对发酵何首乌中的主要化学成分进行初步鉴定,为阐明发酵何首乌的药理作用打下良好的基础。方法:采用HPLC梯度洗脱的方法测定了不同何首乌的特征图谱,计算它们的相似度并对发酵何首乌中主要色谱峰进行了初步鉴定。结果:4批何首乌,3批制何首乌及同一来源的何首乌、制何首乌、发酵何首乌特征图谱与其对照特征图谱的相似度均大于0.99。初步鉴定了发酵何首乌中的8个化合物。结论:4批何首乌、3批制何首乌之间亲缘关系可能较近。发酵何首乌与同一来源的何首乌、制何首乌之间相关性较好,化学成分总体相似,未发现新成分。

Abstract:Objective:To compare the differences of chemical constituents in different Radix Polygoni Multiflori by preliminary studying their specific chromatograms and to identify the primary chemical constituents in fermented Radix Polygoni Multiflori by HPLC-TOF/MS to lay the root for clarifying pharmacological actions of fermented Radix Polygoni Multiflori.Methods: The specific chromatograms of different Radix Polygoni Multiflori were obtained by RP-HPLC with gradient elution.Their similarities were calculated and the primary peaks of fermented Radix Polygoni Multiflori were identified.Results: The similarities of four batches of Radix Polygoni Multiflori,three batches of Radix Polygoni Multiflori Preparata and Radix Polygoni Multiflori from the same origin were all more than 0.99.Eight peaks in Radix Polygoni Multiflori after fermentation were identified.Conclusions: The genetic relationships of the four batches of Radix Polygoni Multiflori and the three batches of Radix Polygoni Multiflori Preparata were likely to be intimate.The correlations of Radix Polygoni Multiflori from the same origin were better and their chemical constituents were similar as a whole.No new constituents were found.
