摘    要:目的鉴定大鼠脑出血(intracerebral hemorrhage,ICH)后的差异蛋白,分析它们在ICH中的病理生理机制,为进一步筛选特异性功能相关蛋白和药物靶点提供线索。方法制作大鼠ICH模型,应用双向凝胶电泳和基质辅助激光解析/电离飞行时间串联质谱技术和Western Blot法,鉴定大鼠ICH后72 h脑组织和正常脑组织的差异蛋白并进行验证。结果在18次重复实验中,共鉴定出39种差异蛋白,其中上调蛋白15种、下调蛋白24种。应用Western Blot法对其中2种关键性蛋白进行了验证。这些差异蛋白涉及细胞骨架、能量代谢酶类、细胞凋亡等方面。结论细胞骨架蛋白与ICH的发病机制密切相关,为探索ICH的病理生理过程和治疗靶点提供关键信息和理论依据。

Quantitative proteomic analysis of rat brain tissue at 72 hour after intracerebral hemorrhage

Abstract:ObjectiveTo identify the differentially expressed proteins in rat intracerebral he morrhage (ICH) and analyze the related physiopathologic mechanism to pave the way for screening special function related proteins and drug targets. MethodsWe made a rat ICH model and then used two dimensional gel electrophoresis (2 DE) and matrix assisted laser desorption ionization time of flight/time of flight mass spectrometry (MALDI TOF/TOF MS) to identify the differentially expressed proteins between the 72 hour after ICH group and the normal control group, and then used Western Blot to validate. ResultsIn total, 39 proteins differentially expressed were identified in 18 replicated experiments, among which 15 proteins are up regulated and 24 proteins were down regulated. Then, 2 key proteins were validated by Western Blot. These differentially expressed proteins include energy metabolic enzymes, antioxidants proteins, apoptosis related proteins cytoskeleton proteins and other proteins. ConclusionCytoskeleton proteins were closely related to pathogenesis of ICH. These may provide the key information and theoretical clue for exploring the ICH physiopathologic process and would be potential therapeutic targets in ICH.
