丹参对大鼠血管平滑肌细胞迁移的影响

摘    要:观察丹参对血管平滑肌细胞迁移的作用,并探讨其作用机理。本研究用羟基脲抑制血管平滑肌细胞增殖后,观察丹参对血管平滑肌细胞迁移、迁移相关基因表达及能量转换的影响。发现丹参对新生小牛血清诱导的血管平滑肌细胞迁移具有显著抑制作用;Northern杂交结果表明丹参可抑制与细胞迁移相关的骨桥蛋白基因表达;肌酸激酶活性分析显示丹参可显著降低该酶活性。上述结果提示:丹参抑制血管平滑肌细胞迁移的作用与抑制骨桥蛋白基因表达和减缓能量转换有关。

Abstract:In order to investigate the effects and the mechanisms of Salvia Miltiorrhiza Bunge(SMB) on the vascular smooth muscle cell (VSMC) migration, the behaviors of VSMC, including migration, migration related gene expression and energy transmission, were observed. It was shown in the result that the migration of VSMC induced by newborn calf serum can be suppressed by SMB. The result of northern blot suggested that the cell migration related osteopontin gene expression can be suppressed by SMB. It was observed that the CK activity can be reduced remarkably by SMB. These results proposed that the inhibition of SMB on VSMC migration is associated with the decrease of osteopontin gene expression and energy transmission.
