^99Tc—MDP对兔实验性葡萄膜炎的治疗作用

摘    要:目的探讨^99锝-亚甲基二膦酸盐(^99Tc-MDP,云克)对兔实验性葡萄膜炎的治疗作用。方法实验兔22只随机分为生理盐水组、地塞米松组、云克组。ELISA法测定实验性葡萄膜炎兔动物模型造模前、造模后5d、给药治疗后10d、停药后5d、4个不同阶段白细胞介素(IL-2)、干扰素(IFN-γ)、肿瘤坏死因子(TNF-α)的血清质量浓度;浊度法及细胞技术池测定不同治疗阶段房水蛋白及房水细胞计数,于不同阶段摘除眼球行组织病理学观察。结果与生理盐水组比较,地塞米松组及云克组均可使兔血清IL-2、IFN-γ、TNF—α水平和房水细胞计数、房水蛋白质量浓度降低,差异均有统计学意义(P〈0.05),地塞米松组与云克组2组间差异无统计学意义(P〉0.05)。地塞米松组及云克组在治疗后眼球的组织病理学显示炎症较治疗前减轻。结论云克在一定程度上有降低兔实验性葡萄膜炎血清IL-2、IFN-γ、TNF-α质量浓度及房水细胞计数、房水蛋白质量浓度以及减轻眼球炎症的作用。

The effect of 99Tc-MDP on experimental autoimmune uveitis in rabbits

Abstract:Objective Animal experiments found that ^99Tc-MDP, an artificial trace element and MDP, can decline immune-regulating level, so it has been used in the treatment of some diseases associated with auto-immune dysfunction. The aim of this study was to investigate the impact of ^99Tc-MDP on the treatment of experimental autoimmune uveitis(EAU). Methods The rabbits EAU models were created by injecting 2% bovine serum albumin(2.5 mL) initially into vitreous and secondly via marginal ear vessels in 21 healthy female white rabbits. The normal saline solution(NS) ,dexamethasone and ^99Tc- MDP was injected via marginal ear vessels in model animals respectively for 10 days. The animals were randomly divided into normal saline control group, dexamethasone administration group,^99Tc-MDP administration group. The interleukin (IL-2) , interferon (IFN-γ),tumor necrosis factor (TNF-α) in serum of rabbits were detected by ELISA assay. Turbidimetric method and cell technology pool method were applied to detect the content of cells and concentration of protein in aqueous humor of rabbits in different time. Eyeballs of experimental animals were enucleated in the different stages for pathologic examination. Results The inflammation score of anterior chamber was 0 before injection of bovine serum albumin, and the highest inflammation score was seen in the 5th day after injection of bovine serum albumin. The inflammation score was gradually decreased after administration of ^99Tc-MDP and dexamethasone in comparison with before use of medicion ( P 〈 0.05, P 〈 0.01 ). Compared with NS group, the levels of IL-2, IFN-γ, TNF-α in rabbit serum was significantly declined in various time points( P 〈 0.05, P 〈 0.01 ). The number of cells and concentration of protein in aqueous humour of rabbits were significantly lower than that of NS group (P 〈 0.05 ) , and no significant difference was found between dexamethasone group and ^99Tc-MDP group( P 〉 0.05 ).
