银杏内酯对内毒素家兔肠动力紊乱调整作用的实验研究

摘    要:实验在清醒禁食家兔通过记录小肠平滑肌电活动的变化观察了银杏内酯A(Ginkgolide A)时内毒素所致小肠运动功能损害的保护作用。实验动物随机分成对照组、内毒素致病组和银杏内酯预处理组。分别记录用药前后和组别之间小肠移行性综合肌电(IMC)周期、各时相时程以及慢波和快波变化情况。发现内毒素30μg/kg静脉注射可引起小肠肌电活动明显抑制和紊乱,IMC中断并诱发出间断性不规则高峰电活动;银杏内酯A 5mg/kg的预处理可显著压抑(P<0.01,n=12)内毒素所致的这一毒性反应,可使IMC周期恢复提前,IMC的各时相时程以及慢波频率和幅度接近于对照水平而不规则高峰电活动时间缩短。结果表明,银杏内酯可通过强烈抑制内毒素介导的小肠肌电紊乱调整平滑肌运动功能。

Adjust Effects of Ginkgolide on Endotoxin-induced Intestinal Motility Function Disturbances in Conscious Rabbits
Abstract:The protection effect of Ginkgolide A on endotoxin - induced small intestinal motility dysfunction Was investigated in rabbits. Thirsty rabbits were randomly assigned to control, endotoxin (Escbericbia coli Lipopolysaccharide, LPS) and Ginkgolides group. Two pairs of electrodes were implanted in the duodena -jejunum , and after seven days, intestinal myoelectric activity was continuously recorded in the conscious , fasted condition. The intravenous administration of Endotoxin(30μg/kg)resulted in an absence of the interdigestive myoelectric complex (IMC) for 108. 5±37. 4min and a decrease on the frequency and amplitude of slow wave re-spectively( P <0. 05 versus control). After a quiescent period,there was a period of irregular spiking burst activity. Previous IV administration of Ginkgolide A(5mg/kg) can significantly reduced the myoelectric activity alterations induced by endotoxin( P <0. 01, n = 12). The duration of irregular spiking of activity was shorted and dysrhythmia index was reduced significantly from 50. 1±17. 53% to 15. 6±4. 5%. The time of IMC cycle recovery was in advance. The phase in IMC cycles and slow wave frequency were also close to control during Ginkgolide. The result showed that Ginkgolide,a specific platelet-activating factor(PAF) receptor antagonist, reduced markedly the intestinal motility dysfunction by endotoxin - induced in rabbits.
