摘    要:目的:探讨大鼠运动区脑片器官型培养的方法及其皮层第V层锥体细胞的鉴定。方法:利用出生后1天的乳鼠大脑活组织切片建立脑片的器官型培养模型,并用抗非磷酸化神经丝单克隆抗体SM-32对皮层锥体细胞加以鉴定。结果:脑片体外生长良好,形态完整,皮层锥体细胞数目恒定,并可存活1个月以上。结论:大鼠脑片的器官培养技术为研究大脑生理、病理改变及神经保护提供了有效的方法。

Rat brain motor area slice organotypic culture and the identification of pyramidal cell

Abstract:Objective: : To establish a method of brain slice organotypic culture for neonate rats and provide a method of identification of pyramidal cells m layer V. Methods. Organotypic brain slice cultures were prepared using brain from 1-day-old rat pups. Pyramidal cells' survival was evaluated by culture morphology and nonphosphorylated neurofilament H (SMI-32) immunohistochemistry staining. Results: The brain explants can be maintained in culture for more than 1 month with excellent organotypic cellular organization and a stable population of cortical pyramidal cells in layer V. Conclusion: Organotypic cultures provide an effective method to study the physiological, pathological changes and neuroprotection of brain.
