调脂降糖片对2型糖尿病大鼠胰岛素抵抗的干预作用

目的观察调脂降糖片对2型糖尿病大鼠胰岛素抵抗(insulin resistance,IR)的干预作用。方法采用一次性腹腔注射小剂量链脲佐菌素(STZ)30mg/kg体重配合高脂饲料喂养建立2型糖尿病胰岛素抵抗大鼠模型。用调脂降糖片干预治疗,以二甲双胍片作为阳性对照。8周后采用血糖钳技术检测大鼠葡萄糖输注率(GIR),测定大鼠空腹血糖(FBG)、空腹胰岛素(FINS)、血清游离脂肪酸(FFAs)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C),并计算高密度脂蛋白胆固醇/总胆固醇比值(HDL-C/ TC)、胰岛素敏感指数(ISI)。对大鼠的胰岛素敏感性和脂代谢情况进行评估。结果与模型对照组相比,调脂降糖片治疗后大鼠TC、TG、HDL-C/TC、FINS、FFAs显著下降,而ISI、GIR明显增加,其作用与二甲双胍片基本相似。结论调脂降糖片能够增加2型糖尿病大鼠胰岛素敏感性,并改善糖、脂代谢异常。

Intervention Effects of Tiaozhi Jiangtang Tablet on Insulin Resistance in Rats with Diabetes Mellitus Type 2

Objective To observe the effect of Tiaozhi Jiangtang Tablet(TJT)on insulin resistance(IR)in rats with diebetes mellitus type 2.Methods The model rots of diebetes mellitus were induced by intraperitoneal injection of streptozotocin(30mg/kg)and feeding with high lipid forage.The rats in the TJT group were treated with TJT and those in the mefformin group treated with mefformin as positive controls.The glucose infusion rate (GIR)was detected by glucose clamp technique after treatment for 8 weeks.At the same time,fasting blood glu- cose(FBG),fasting insulin(FINS),free fatty acids(FFA),total cholesterol(TC),triglyceride(TG),and high- density lipoprotein cholesterol(HDL-C)were measured respectively,and insulin sensitivity index(ISI)and HDL- C/TC calculated.The changes of insulin sensitivity and lipid metabolism were evaluated.Results TC,TG,HDL- C/TC,FINS,and FFA significantly reduced in the TJT group as compared with those in the control group,while ISI and GIR significantly increased,the effects of TJT were similar to those of mefformin.Conclusion TJT is effective in increasing insulin sensitivity and improving glucose and lipid metabolisms in rats with diebetes mellitus type 2.
