摘    要:目的]研究基质金属蛋白酶-9(MMP-9)在大鼠光气吸入性肺损伤中的作用机制。方法]利用三光气在N,N-二甲基甲酰胺作用下分解生成光气的方法,染毒柜内通入浓度为8.33mg/L的光气。40只SD大鼠随机分为染毒2、4、6、8h组和空气对照组。测定肺组织湿干比和支气管肺泡灌洗液中蛋白含量并观察肺组织病理学改变,免疫组化方法测定肺组织中MMP-9的含量。结果]MMP-9在大鼠光气吸入性肺损伤的模型中表达较对照组明显增加(P〈0.05),并与观察时间延长、肺损伤的加重有相关性。结论]MMP-9在光气吸入性肺损伤时表达增加,并发挥加重肺损伤的作用。

Abstract:Objective] To investigate the role of matrix metalloproteinase-9(MMP-9)in the pathogenesis of acute lung injury induced by phosgene.Methods]Phosgene was produced by the decomposition of BTC in the catalysis of DMF,and the concentration of phosgene in the intoxication chamber was 8 mg/L.40 SD rats were randomly separated into 5 groups,the control group and the experiment groups(2,4,6,8 h after phosgene inhalation).Experiment rats were sacrificed at 2,4,6,8 h after phosgene inhalation.Wet to dry weight ratio of the lung(wet/dry)and the contents of protein in bronchoalveolar lavage fluid(BALF) were recorded,histopathology of the lung was observed under the light microscope,the expression of MMP-9 in lung tissue was verified by immunohistochemical analysis.Results]Acute lung injury caused by phosgene was accompanied with increased expression of MMP-9 in lung tissues(P<0.05),which was correlated with the elongation of time and aggravation of lung injury. Conclusion]MMP-9 plays an aggravating role in the development of phosgene-induced acute lung injury in rats.
