Urocortin在胎盘中的分泌与表达

目的:研究Urocortin在胎盘组织产生、分布与分泌的情况,以证实Urocprtin可能由妊娠组织产生并调节子宫-胎盘局部的血管张力。方法:用免疫组化法检测足月妊娠临产及未临产的胎盘、胎膜组织以及剖宫产和非妊娠子宫肌层血管平滑肌细胞中的Urocortin分布。并用细胞培养免疫印迹法检测绒毛膜-蜕膜细胞分泌Urocortin的情况。结果:具有免疫活性的Urocortin分布在蜕膜的间质细胞、滋养层细胞、羊膜上皮细胞和子宫平滑肌内的血管平滑肌细胞;非妊娠子宫血管平滑肌细胞的Urocortin免疫活性明显低于足月妊娠之子宫。培养的蜕膜细胞中48％含有Urocortin,其中41％有基础状况下的分泌。结论:在人类妊娠过程中Urocortin由蜕膜细胞产生并分泌,为Urocortin由妊娠组织产生并局部调节子宫-胎盘血管张力的学说提供了实验基础。

The secretion and expression of Urocortin in placenta

ve:To study the producing, distribution and secretion of Urocortin in pla-centa , and verify the hypothesis that Urocortin produced in the pregnant tissues and locally regulated the utero-placental vascular tone during pregnancy. Methods: The distribution of Urocortin in human placenta, fetal membranes and uterine tissues at term in the presence and absence of labor, and in u-terine vascular smooth muscle cells at term pregnancy and non-pregnancy were examined by immuno-histochemistry method. Subsequently, Urocortin secretion from chorio-decidual cells in vitro was test-ed by an immunoblot technique.Results: Urocortin was observed in the decidual stromal cells, syn-cytiotrophoblast, amnion epithelium cells and vascular smooth muscle cells of the myometrium at term. Staining intensity for Urocortin in vascular smooth muscle cells of the myometrium at term was stronger than that at non-pregnancy. Positive staining for Urocortin was observed 48% in cultured chorio-decidual cells and with 41% of these cells secreting Urocortin under basic conditions. Con-clusions: Urocortin is produced by the decidua cells during human pregnancy. These data are consis-tent with the hypothesis that Urocortin is produced locally by the pregnant tissues and may act to reg-ulate utero-placental blood flow.Urocortin;Corticotropin-releasing hormone;Pregnancy;Placenta;Fetal membranes
