小鼠急性柯萨奇B_3病毒性心肌炎电生理的表现

摘    要:用常规细胞内微电极技术及实时微机数据处理系统,对柯萨奇B_3病毒(CB_3V)感染的BALB/c小鼠(早期7d、14d)作心肌电生理参数及动作电位波形研究。发现在病毒感染早期,心肌细胞的静息电位(RP)、动作电位振幅(APA)、超射(Os)、O相最大上升速率(Vmax)均数值除感染7d组Os值无差异,其余均显著低于同期正常对照组:而动作电位时程APD50和APD90均数值除感染14d组APD90无差异外,均显著延长。动作电位异常波形多达十种,占感染组的65%以上。提示CB_3V感染对心肌造成的损害,可产生多种心电活动异常,并成为形成各种形式心律失常的基础。

Abstract:Acute Coxsackie B3 virus (CB3V) myocarditis experimentally produced in 7 and 14 days old BALB/c mice. Myocardial virus titer, neutralizing antibody, and electrical activity of the right ventricular myocardium were examined by standard intracellular microelectrode techniques and a real-time microcomputer data processor srstem.CB3V was isolated from the infected myocardium on the 7th day, but no virus could be isolated on the 14th day. CB3V neutralizing antibody was detected in the sera of both infected groups. Decrease of rest potential (RP), maximum upstroke rate (Vmax), overshoot(Os), amp'itide of action potential (APA), and prolongation of action potential duration (APD50 and APD90) occured in both two infec ted groups. Meanwhile, 10 types of abnormal action potentials were abserved frequently throughout the entire experiment. Our findings would be useful in tsudy of the pathogenesis and treatment of multiform arrhythmias in viral myocarditis.
