何首乌水提液对去卵巢大鼠骨组织的动态影响

摘    要:目的:观察何首乌水提液对去卵巢大鼠骨组织的动态影响。方法:把64只6月龄SD雌性大鼠随机分为4组,即假手术组、卵巢切除组、何首乌组、尼尔雌醇(E2)组,每组16只。各组大鼠分别于开始给药后第15日、第30日将各组的半数动物处死,测定骨羟脯氨酸(HOP)、骨钙、骨磷、血清及骨碱性磷酸酶(ALP)、血清及骨抗酒石酸酸性磷酸酶(TRAP)并做了骨组织的病理HE染色。结果:在两个阶段的治疗中,何首乌组与卵巢切除组比较,骨有机质和骨矿物质均有显著性升高;骨组织病理形态改善;TRAP活性变化没有出现明显改变(P〉0.05),而ALP活性出现明显的变化。骨组织ALP比血清ALP的统计学差异更大,并且以上指标在第二阶段较第一阶段的统计学差异更为显著。结论:何首乌水提液可能主要通过增强ALP活性,抑制骨胶原、骨钙、骨磷的丢失,从而抑制去卵巢大鼠的骨质疏松。

Abstract:Objectiv:Observing bone dynamic effect on the ovariotomized rat treated with extract of Polygonum multifloruma.Methods:sixty-four female six months SD rats were divided into 4 groups randomly(16 rats/group):sham ovariectomized group(Sham),ovariectomied group(OVX),ovariectomied+Polygonum multifloruma group(HSW),ovariectomized+Nilestriol group(E2).Thirty days after operation,HSW or E2 group were given Polygonum multifloruma or Nilestriol respectively,Sham and OVX groups are given distilled water once a day.Half animals of each group were killed on 15th day and 30th day respectively.Serum,femurs and tibias were collected.And then following tests were made:serum and bone alkaline phosphatase(ALP),serum and bone tartrate-resistant acid phosphatase(TRAP),and bone hydroxyproline(HOP),calcium(Ca),phosphorus(P),bone morphological features of femur.Results:Compared with OVX group,Bone organic and mineral were remarkably increased in HSW and E2 groups during the 2 terms.Bone morphological features of femur were improved.Bone TRAP activity had no obvious changes in E2 group(P>0.05).Bone ALP activity had obvious changes in HSW group(P<0.05).In addition,all the markers on 2nd term had more obvious difference than the 1st term on statistics.Conclusion:Polygonum multifloruma could promote bone ALP activity to inhibite the lost of bone organic and mineral content,and improve bone morphological features of osteoporotic femur in ovariectomied rats.
