结肠癌患者MMP-9的表达及临床意义

目的测定基质金属蛋白酶(MMP-9)在结肠癌患者血清中的水平及组织中的表达,探讨其与结肠癌恶性程度、浸润、转移的关系。方法采用ELISA法及免疫组织化学SP法检测42例结肠癌患者血清MMP-9水平及结肠癌组织中MMP-9表达,分析与结肠癌恶性程度、浸润、转移的关系。结果1.结肠癌患者血清MMP-9水平显著高于对照组(P<0.01),Dukes C期、D期血清MMP-9水平显著高于A、B期(P<0.01),低分化者血清MMP-9显著高于高、中分化者(P<0.05)。2.结肠癌MMP-9蛋白表达阳性率高于正常结肠组织,但无统计学差异(P>0.05),而Dukes分期C D期阳性表达率显著高于A B期(P<0.05),有淋巴结转移者阳性表达率显著高于无淋巴结转移(P<0.05)。结论MMP-9与Dukes分期、淋巴结转移有关,具有重要的临床意义。

Expression of Matrix Metalloproteinase-9 Protein in Colonic Carcinoma and Its Significance

Objective To analyze the correlation of matrix metalloproteinase-9(MMP-9) expression and the serum levels of MMP-9 with the pathological factors in colonic carcinoma and to investigate the clinical significance of MMP-9 in the occurrence of colonic carcinoma.Methods The expression and the serum levels of MMP-9 were measured by immunohistochemistry and ELISA in the resected specimens from 42 patients with colonic carcinoma and 40 controls and their relations with clinicopathologic factors were analyzed.Results 1.Serum levels of MMP-9 were significantly higher in the colonic carcinoma patients than those of controls(P<0.01).The levels were associated with tumor differentiation and Dukes stage.Patients with poor differentiated and Dukes C or D stage tumors had significantly higher levels MMP-9 than those of patients with well differentiated and Dukes A or B stage tumor(P<0.01).2.The positive rate MMP-9 protein expression was no significant difference between colonic carcinoma(P>0.05),while the positive rates of MMP-9 protein expression were significantly correlated with the Dukes stage and lymph node metastasis(C D stage vs A B stage,P<0.05,metastasis vs non metastasis,P<0.05).Conclusion Over expression of and elevated MMP-9 play important roles in the carcinogensis and metastasis of colonical carcinoma.
