谷氨酰胺、精氨酸对小肠上皮细胞生长的促进作用

摘    要:探讨谷氨酰胺、精氨酸,单独以及两者联合使用对原代培养的小肠上皮细胞生长和分化的影响。方法:实验分谷氨酸胺、精氨酸、谷氨酰胺加精氨酸三组,于小肠上皮细胞原代培养中加入不同剂量的药物,测定细胞的生长和ALP、CK活性。结果:单一谷氨酰胺或精氨酸促进肠上皮细胞生长和减少细胞死亡的作用明显优于两者联合用药效果,谷氨酰胺促进细胞分化,精氨酸使细胞分化降低,两者联合亦使细胞分化降低。结论:单一使用谷氨酰胺或精氨酸能促进肠上皮细胞生长,两者联合后其肠营养作用下降。这为临床合理地选择和搭配各种肠营养因子提供理论依据。

Abstract:Objective : In considering new forms of nutritional support for the epithelial cells of small intestine, we designed this study to investigate glutamine , arginine, and the combined effects of glutamine and arginine in vitro on the proliferation and differentiation of small intestinal epithelium, and characterize the interactions between them on the intestinal mucosa. Methods: Two groups received glutamine and arginine at different levels alone. The third group received both glutamine and arginine at the same levels. Cell proiferation was determined by MTT colorimetric assay. Alkaline phosphatase (ALP)and creatine phosphokinase (CK) activities of epithelial cell were also measured. Results: It was showed that either glutamine or arginine had a stronger trophic effect on the epithelial growth of small intestine in primary culture. However, the combined effects of glutamine and arginine on the proiferation of small intestinal epithelium in a manner less than that caused by either agent alone. Also we found that besides the proliferation, both glutamine and arginine were able to decrease the appearance of epithelial death.Conclusion: Either glutamine or arginine had a stronger effect on the epithelial growth of small intestine in primary culture. On the contrary, their combined effects were in a manner less than that caused by either agent alone. The results were very important for specific nutrients to be chasen in comination to decrease the mucosal atrophy that commonly occurs after disuse or disease.
