米非司酮与利洛司酮对异位子宫内膜间质细胞凋亡的影响

摘    要:目的 探讨米非司酮和利洛司酮对异位子宫内膜间质细胞凋亡形态学和凋亡率的影响。方法 应用光学显微镜观察细胞生长状态 ;应用电子显微镜观察凋亡细胞的超微结构 ,应用 TUNEL法观察细胞凋亡率。结果 两种抗孕激素以浓度 1× 10 - 6 mol/ L、培养 4 8小时 ,异位子宫内膜间质细胞开始出现早期细胞凋亡变化 ,随作用时间延长和浓度增加 ,细胞凋亡形态学变化更趋明显 ,细胞凋亡率增加 ,利洛司酮诱导细胞凋亡作用大于米非司酮。结论 抗孕激素米非司酮和利洛司酮以时间和剂量依赖性方式促进异位子宫内膜间质细胞的凋亡 ;利洛司酮诱导异位内膜细胞凋亡的作用高于米非司酮

Abstract:Objective To investigate the effects of antiprogestin mifepristone and lilopristone under different dose and time on the apoptosis rate and morphology of the ectopic stromal cells in vitro.Methods The morphology of ectopic stromal cells of endometriosis was observed by optical microscopy and the super microstructure of apoptosis cells were observed by electronic microscopy.The apoptosis rate of ectopic stromal cells was determined by TUNEL analysis.Results The early morphological changes of cell apoptosis in ectopic stromal cell appeared when the antiprogestins concentration was 10 -6 mol/L after culturing for 48 hours.The apoptosis morphological changes and apoptosis rate of ectopic stromal cell became more typical following the increased of the time and dose of antiprogestins.The effect of lilopristone was higher than that of mifepristone in reducing apoptosis ectopic stromal cell in vitro.Conclusions Antiprogestins mifepristone and lilopristone could promote apoptosis of ectopic stromal cells in time-dose dependent fashion in vitro.The effect of lilopristone inducing apoptosis of ectopic stromal cell;higher than that mifepristone in vitro.
