肺腺癌组织中CPNE1蛋白的表达及临床意义

目的探讨Copine 1(CPNE1)蛋白在肺腺癌组织中的表达及意义。方法采用免疫组化SP法检测CPNE1蛋白在186例肺腺癌组织及30例正常肺组织中的表达;采用RT-PCR技术检测肺腺癌组织及正常肺组织中CPNE1 mRNA的表达。结果在186例肺腺癌组织中68例CPNE1低表达(36.56%),118例呈CPNE1高表达(63.44%),CPNE1在正常组织中均呈低表达。相关性分析结果显示,CPNE1表达与肺腺癌分期、远处转移有关(P<0.05),与患者年龄、性别、肿瘤大小等无关。RT-PCR结果显示,肺腺癌组织中CPNE1的表达量显著高于正常肺组织。结论CPNE1蛋白在肺腺癌组织中的表达较正常肺组织高,且与淋巴结转移、临床分期有关。CPNE1蛋白表达与肺腺癌的发生、发展及预后密切相关,有望成为肺腺癌预后评估的重要指标。

Expression of Copine 1 protein and mRNA in human lung adenocarcinoma and its clinical significance

Purpose To investigate the expression of Copine 1(CPNE 1)protein and mRNA in human lung adenocarcinoma and to explore its role and significance in the progression of lung adenocarcinoma.Methods The expression of CPNE1 in 186 lung adenocarcinoma patient tissues and 30 normal lung tissue were initially investigated by immunohistochemical staining,the CPNE1 mRNA expression level was also determined by real-time PCR.Results Among the 186 cases of lung adenocarcinoma,118 cases showed high expression of CPNE1(63.44%),68 cases showed low expression of CPNE1(36.56%),and low CPNE1 expression was found in 30 normal tissue.CPNE1 expression level was positively associated with stage,lymph node status,and distant metastasis(P<0.05),but unrelated to the age,sex and tumor siza(P>0.05).RT-PCR showed that the expression of CPNE1 in lung adenocarcinoma tissues was significantly higher than normal tissues.Conclusion CPNE1 expression level in lung adenocarcinoma tissues is significantly higher than normal tissues and it is positively associated with lymph node status and clinical stage.CPNE1 expression is closely related to the occurrence,development and prognosis of lung adenocarcinoma,which may become a new marker for the prognosis of lung adenocarcinoma.
