Caveolinl基因敲除对于小鼠脑缺血神经功能及脑梗死面积恢复的影响

摘    要:目的:评价小窝蛋白caveolinl在MCAO术后脑损伤中起的作用。方法:将C57/BALB/C小鼠分为正常假手术组和正常模型组,caveolinl基因敲除小鼠分为基因敲除假手术组和基因敲除模型组,每组20只.对两模型组小鼠行MCAO术造成脑缺血损伤模型。分别于造模术后第l、7天行TTC染色评价梗死面积,称大脑干湿重评价脑水肿程度,并进行神经功能评分。结果:与正常小鼠比较,caveolinl基因缺失小鼠的损伤变化增强,恢复速度明显减缓、结论:caveolinl在MCAO脑损伤中可能起保护作用和促恢复作用。

Effect of caveolin - 1 gene knockout on recovery of neurological function and area of cerebral infarction in mice with cerebral ischemic injury

Abstract:Objective : To evaluate the role of caveolin - 1 in cerebral injury induced by middle cerebral artery occlusion (MCAO). Methods :C57/BALB/c mice were divided into normal sham- operation group (n = 20) and normal model group (n = 20) ,while cave- olin - 1 gene knockout mice were divided into gene knockout sham - operation group ( n = 20) and gene knockout model group ( n = 20). The mice in model groups were treated by MCAO to establish a model of cerebral ischemic injmT. At 1 and 7 days alter the model was established,TFC staining was used to measure the area of cerebral infarction,the dry and wet weights of lhe brain were determined to evaluate the severity of cerebral edema,and the neurological score was calculated. Resuhs:Compared with normal mice,caveolin -1 gene knockout mice had increased cerebral injury and significantly slowed recovery of neurological function. Conclusion:Caveolin - 1 may play a protective role and promote the recovery of neurolngical function in MCAO - induced cerebral injury.
