摘    要:目的探讨谷胱甘肽-S-转移酶(rSj26GST)多克隆抗体诊断日本血吸虫病的应用价值。方法以抗rSj26GST多克隆抗体力捕获并检测抗体,建立双抗体夹心斑点免疫金测定技术,用于检测日本血吸虫循环抗原,并以斑点免疫金测定法和ELISA检测抗体作对照。结果3种方法检测急性血吸虫病的敏感性均为100%;对慢性血吸虫病的敏感性分别为76.4%、98.1%和96.2%;特异性分别为95.0%、98.0%和96.0%。结论rSj26GST抗原对日本血吸虫病具有诊断价值,可望有较好的应用前景。

Abstract:Objectives To study the value of diagnosis for schistosomiasis japonica by gold immuno-assay with polyclonal antibodies against recombinant Schistosoma japonicum 26 kilo-dalton glutathione s-transferase (rSj26GST) antigen. Methods Polyclonal antibodies against rSj26GST were used as capture and label antibodies, and double antibodies sandwich dot immuno-gold filter assays (S-DIGFA) was developed for detecting circulating antigen of Schistosoma japonicum. Two assays, dot immuno-gold filter assay (SEA-DIGFA) and ELISA(SEA-ELISA) detecting antibodies against SEA, were compared with S-DIGFA. Results The positive rates of these three methods were all 100% for acute schistosomiasis. And for chronic schistosomiasis, the positive rates were 74. 6%, 98. 1 % and 96. 2%,respectively. The negative rates of these three methods for normal persons were 95. 0%,98. 0% and 96. 0%, respectively. Conclusion The method with polyclonal antibodies against rSj26GST showed potential value for diagnosis of schistosomiasis japonica.
