高危或极高危原发性高血压患者外周血单核细胞趋化因子-1基因的表达

摘    要:目的:探讨高危或极高危原发性高血压(EH)患者外周血单核细胞趋化因子-1(MCP-1)表达,为寻找高危或极高危EH患者的治疗手段提供理论依据。方法:运用酶联免疫吸附(ELISA)法测定高危或极高危EH患者65例(EH1组)、低或中危EH患者35例(EH2组)和30例正常对照者外周血MCP-1蛋白含量,用逆转录聚合酶链反应(RT-PCR)法检测外周血单个核细胞中MCP-1 mRNA的表达。结果:与对照组比较,EH1组和EH2组外周血MCP-1蛋白含量和单个核细胞中mRNA表达均显著升高,具有统计学差异(P〈0.05orP〈0.01);与EH2组比较,EH1组患者外周血MCP-1蛋白含量和单个核细胞中mRNA表达均显著升高,具有统计学差异(P〈0.01)。结论:外周血单个核细胞中MCP-1可能参与了高危或极高危EH的病理生理过程,外周血单个核细胞的MCP-1 mRNA表达上调可能是高危或极高危EH患者外周血MCP-1蛋白含量增高的机制之一。

Abstract:Objective:To observe the level changes of protein and mRNA of monocyte chemoattractant protein-1 (MCP-1) in peripherial blood of patients with essential hypertension (EH) in high or very high risk grades. Methods:Concentrations of circulating MCP-1 of 65 EH patients with high or very high risk (group EH1),35 EH patients with low or moderate risk (group EH2),and 30 normotensive healthy people (group C) were tested with enzyme linked immunosorbent assay (ELISA),and expression of MCP-1 mRNA in peripherial blood mononuclearcells of the subjects were determined with RT-PCR. Results:MCP-1 protein level and intra-mononuclearcell expression of MCP-1 mRNA in peripherial blood were significantly higher in groups EH1 and EH2 than in group C (P0.05 or P0.01),and were significantly higher in group EH1 than in group EH2 (P0.01). Conclusions:MCP-1 in peripherial blood might participate in the patho-physiology process of EH patients in high or very high risk grades; Expression increase of mRNA MCP-1 may be one of the mechanisms of protein MCP-1 increasing in peripherial blood in these patients.
