摘    要:目的观察Ad-huVEGF121体外表达效率及其表达产物的生物学活性。方法重组腺病毒Ad-huVEGF121转染NIH3T3细胞,应用RT-PCR和免疫组织化学技术检测VEGF121的表达,表达产物在兔股骨骨膜下注射,观察其生血管活性。结果转染后NIH3T3细胞持续表达VEGF121 10d左右,骨膜下注射后新生血管数量显著多于对照组。结论我们构建的AD-huVEGF121能在真核细胞内高效表达目的基因,其表达的蛋白具有促进血管生成的生物学活性。

Abstract:Objective To observe the expression efficiency of Ad-huVEGF121 in NIH 3T3 cell and the angiogenesis activity of its expression product. Methods The Ad-huVEGF121 was transfected to NIH 3T3 cell, The expression of huVEGF121 was examined by RT-PCR and immunohistochemical techniques, angiogenesis activity was observed by CD105 immunohistochemical techniques after the expression product was injected into rabbit subperiosteum of femur. Results The expression of huVEGF121 was detected in NIH 3T3 after transfected with the huVEGF121 gene for 10d, The subperiosteum of femur that was transfected with the huVEGF121 gene showed a significant increase in angiogenesis after gene transfection than the control groups. Conclusions The gene of Ad-VEGF121 can express in eukaryocyte and its expression product possess angiogenesis activeity.
