加味滋生青阳片对自发性高血压大鼠肾脏的保护作用及机制研究

目的观察加味滋生青阳片对自发性高血压大鼠(SHR)血压、高敏C反应蛋白(hs-CRP)、白细胞介素-1β(IL-1β)、白细胞介素-10(IL-10)、肿瘤坏死因子-α(TNF-α)及肾动脉病理学的影响,探讨其对SHR肾脏保护作用及机制。方法将36只12周龄雄性SHR按照完全随机原则分为SHR模型组、中药组、替米沙坦组,同时设置12只WKY大鼠为空白对照组。其中中药组予加味滋生青阳片灌胃,替米沙坦组予替米沙坦灌胃,模型组及空白对照组予相同体积的无菌用水灌胃,连续干预8周。监测各组大鼠血压;用ELISA法测定血清hsCRP、IL-1β、IL-10、TNF-α的水平;HE染色进行肾动脉病理学观察。结果中药组、替米沙坦组给药4、6、8周后血压明显下降,差异具有统计学意义(P<0.05或P<0.01),中药组血压与替米沙坦组比较,差异无统计学意义(P>0.05);治疗8周后中药组较模型组血浆hs-CRP、IL-1β、TNF-α均降低,IL-10升高,中药组IL-1β、TNF-α低于替米沙坦组,差异具有统计学意义(P<0.05或P<0.01);替米沙坦组和中药组的肾动脉壁炎症浸润细胞明显减少,弹性纤维排列基本正常,情况明显优于模型组。结论加味滋生青阳片对SHR肾脏具有显著的保护作用,具体机制可能与其降低血压及血浆hs-CRP、IL-1β、TNF-α,升高IL-10相关。

Study on the Protective Effect and Mechanism of Jiawei Zisheng Qingyang Tablets on the Kidney of Spontaneously Hypertensive Rats

Objective:To observe the effects of Jiawei Zisheng Qingyang Tablets on blood pressure,high sensitivity C-reactive protein(hs-CRP),interleukin-1β(IL-1β),interleukin-10(IL-10),tumor necrosis factor-α(TNF-α)in spontaneously hypertensive rats(SHR),and to explore its renal protective effect and mechanism on SHR.Methods:A total of 36 male SHR of 12-week-old were randomly divided into the SHR model group,traditional Chinese medicine(TCM)group and telmisartan group,and WKY rats blank control group with the same number of rats were set up.The rats in the model group and the blank control group were given the same volume of sterile water by gavage for 8 weeks.The levels of hs-CRP,IL-1β,IL-10 and TNF-αin serum were measured by ELISA;the renal artery pathology was observed by HE staining.Results:The blood pressure of TCM group and telmisartan group decreased significantly after 4,6 and 8 weeks(P<0.05 or P<0.01),but there was no significant difference between the TCM group and telmisartan group(P>0.05);after 8 weeks of treatment,the plasma hs CRP,IL-1β,TNF-αin TCM group were lower than those in model group,and IL-10 was increased,while IL-1βand TNF-αin the TCM group were lower than those in telmisartan group,and the differences were statistically significant(P<0.05 or P<0.01);in telmisartan group and traditional Chinese medicine group,inflammatory infiltrating cells in renal artery wall were significantly reduced,elastic fiber arrangement was basically normal,and the situation was significantly better than that of model group.Conclusion:Jiawei Zisheng Qingyang Tablets has a significant protective effect on the kidney of SHR.The specific mechanism may be related to the decrease of blood pressure,the decrease of hs CRP,IL-1β,TNF-αand the increase of IL-10 in SHR.
