血管细胞粘附分子1在分化型甲状腺癌中的表达及临床意义

摘    要:目的:检测血管细胞粘附分子1(VCAM1)在分化型甲状腺癌(DTC)中的表达情况,观察过表达VCAM1甲状腺细胞的侵袭和迁移能力。方法:收集医院收治的60例DTC患者临床资料,按照VCAM1蛋白表达情况将其分为阳性组(39例)和阴性组(21例)。对两组患者DTC组织及癌旁甲状腺组织分别采用实时荧光定量聚合酶链反应(PCR)法、蛋白免疫印迹和免疫组织化学法检测DTC组织及癌旁甲状腺组织VCAM1mRNA和蛋白表达情况,对比DTC组织中不同病理分级、肿瘤大小及预后的蛋白表达阳性及阴性表达情况。采用慢病毒转染法建立过表达甲状腺细胞系FTC-133-VCAM1、空载细胞系FTC-133-control,采用Transwell小室及划痕试验检测并对比FTC-133、FTC-133-VCAM1及FTC-133-control细胞系的穿膜细胞数及迁移率。结果:DTC组织与癌旁甲状腺组织比较,VCAM1mRNA和蛋白相对表达量较高,差异均有统计学意义(t=43.724,t=44.423;P<0.05);DTC组织VCAM1阳性表达构成比为65.00%,高于癌旁甲状腺组织(13.33%),差异有统计学意义(χ2=33.611,P<0.05);阳性组存在淋巴结转移、复发及死亡患者占比分别为33.33%、17.95%和15.38%,显著高于阴性组,差异均有统计学意义(χ2=4.127,χ2=4.065,χ2=3.867;P<0.05);FTC-133-VCAM1细胞与FTC-133、FTC-133-control细胞相比,穿膜细胞数及迁移率较高,差异均有统计学意义(t穿膜细胞数=5.107,t=5.440;t迁移率=14.108,t=13.387;P<0.05)。结论:VCAM1在DTC中异常高表达,且与淋巴结转移及预后关系密切,推测与VCAM1高表达增强癌细胞侵袭及迁移能力有关。

Abstract:Objective:To detect the expression of vascular cell adhesion molecule-1(VCAM1)in differentiated thyroid carcinoma(DTC),and to observe invasion and migration ability of VCAM1 thyroid cells with overexpression.Methods:The clinical data of 60 patients with DTC admitted to hospital were selected.According to the expression of VCAM1 protein,there were 39 cases in the positive group and 21 cases in the negative group.The real-time fluorescence quantification polymerase chain reaction(PCR),Western blot and immunohistochemistry were adopted to detect VCAM1 mRNA and protein expression of DTC tissues and paracancerous thyroid tissues of two groups.The positive and negative expressions of protein in different pathological grades,tumor size and prognosis of DTC tissues were compared.Overexpressed thyroid cell line FTC-133-VCAM1 and no-load cell line FTC-133-control were established by adopting lentivirus transfection.The number of transmembrane cells and mobility of FTC-133,FTC-133-VCAM1 and FTC-133-control cell lines were detected and compared by Transwell chamber assay and scratch test.Results:The relative expression levels of VCAM1 mRNA and protein in DTC tissues were significantly higher than those in paracancerous thyroid tissues(t=43.724,t=44.423,P<0.05).The VCAM1 positive expression in DTC tissues was 65.00%,which was significantly higher than 13.33%in paracancerous thyroid tissues(χ2=33.611,P<0.05).The composition ratios of patients with lymph node metastasis,recurrence and death in VCAM1 positive group were 33.33%,17.95%and 15.38%,which were significantly higher than those in negative group respectively(χ2=4.127,χ2=4.065,χ2=3.867,P<0.05).The number of transmembrane cells and migration rates of FTC-133-VCAM1 cells were significantly higher than those of FTC-133 and FTC-133-control cells,respectively(tNumber of transmembrane cells=5.107,t=5.440,tmigration rate=14.108,t=13.387,P<0.05).Conclusion:VCAM1 has abnormal high expression in DTC,and it is closely related to lymph node metastasis and prognosis.It is speculated that the high expression of VCAM1 may enhance the ability of invasion and migration of cancer cells.
