| 摘 要： | 作者在石蜡组织块作成的电镜切片上,选择需作电镜观察的组织部位,从石蜡块上取下该组织部位,用1％四氧化锇-二甲苯混合液脱蜡再固定,环氧树脂618快速包埋,按电镜常规切片、染色,作成电镜观察用的切片,经电镜观察,效果较为满意,可达到电镜病理诊断要求。本法步骤简单易行,省时,全过程仅需两个半小时。

| Abstract: | A rapid embedding method used inpreparation of paraffin tissue block fortransmission electron microscopy is recom-mended. As to the procedures, selection ofthe tissue area for electron microscopy wasdone first under light microscope, and thecorresponding part was taken off the par-affin tissue block. Then the tissue was de-paraffinised and postfixed in a mixed solu-tion of 1% osmium tetroxide and xylol, andwas embedded in Epox 618. And then ul-trathin section was cut with a glass knife,stained with uranyl acetate and lead tartra-te and finally examined with H600-IVTEM. This method is simple, easy andtimesaving. The whole process takes onlytwo and a half hours with good results. Sothis method can be used for pathologic dia-gnosis. |
