人肺癌组织中C-Ha-ras,C-myc和C-sis基因扩增的研究

摘    要:用同位素α-P32dCTP标记癌基因C-Ha-ras,c-myc和V-sis基因片段,通过点杂交和Southern杂交法检测17例鳞癌、9例腺癌、3例其它类型肺癌组织和9例非肺癌组织中C-Ha-ras,C-myc和C-sis基因扩增的情况。结果发现:在所检测的29例肺癌样品中,10例(58.8%)鳞癌,3例腺癌(33.3%)和2例其它类型癌有C-Ha-ras基因扩增,4例(23.5%)鳞癌,3例(33.3%)腺癌出现C-myc基因扩增,4例(23.5%)鳞癌,1例(11.1%)腺癌和1例其它类型癌有C-sis基因扩增。同时我们也发现有5例肺癌组织(4例鳞癌和11例腺癌)出现两种癌基因的扩增。上述结果提示:C-Ha-ras,C-myc和C-sis基因的扩增与肺癌的发生有密切关系,特别是C-Ha-ras基因扩增在肺鳞癌的形成过程中可能起着重要作用。同时也提示:肺癌的发生与两件癌基因的扩增可能有一定关系。

Abstract:Amplification of C-Ha-ras, C-myc and C-sis oncogene was detected in 29 samples of lung cancer tissues and 9 samples of none lung cancer tissues by dot blotting and southern hybridization. The results showed: C - Ha - ras gene amplification was observed in 10 out of 17 squamous carcinomas (58. 8%) and 3 out of 9 adenocarcinomas (33. 3%); C-myc gene amplification was observed in 4 out of 17 squamous carcinornas(23. 5%)and 1 out of 9 (11. 1 %)adenocarcinomas. And also two oncogene amplificated simultaneously in 4 of squamous carcinomas and 1 of adenocarcinomas. These results suggested that the amplification of the three oncogenes was correlated with the development of lung cancer. Especially the amplification of C-Ha -ras gene may play an important role in the development of squamous cacinomas. And the amplification of two oncogene ndght be correlated with lung cancer.
