白英提取物诱导乳腺癌MCF-7细胞凋亡及其对凋亡相关基因表达的影响

摘    要:目的探讨白英提取物(Solanum lyratum Thunb extract,STE)对乳腺癌MCF-7细胞凋亡及凋亡相关基因survivin和caspase-3表达的影响。方法体外培养MCF-7细胞,以2.5、5、10 mg/L STE处理MCF-7细胞48小时,并以2.5 mg/L顺铂(DDP)作为阳性对照。用MTT法检测细胞抑制率,TUNEL法检测凋亡率,半定量RT-PCR检测survivin和caspase-3 mRNA表达。结果与正常对照组比较,STE各浓度组细胞抑制率显著上升(P<0.001),细胞凋亡率明显升高(P<0.001),caspase-3 mR-NA表达显著上升(P<0.001),survivin mRNA表达明显下降(P<0.05或P<0.01)。结论STE具有诱导MCF-7细胞凋亡作用,其分子机制可能与上调caspase-3及下调survivin基因表达有关。

Abstract:Objective To explore the apoptosis-inducing effect of Solanum lyratum Thunb extract(STE) and the expression of apoptosis associated genes survivin and caspase-3 on human breast cancer MCF-7 cells.Methods MCF-7cells were treated with 2.5,5 and 10 mg/L STE for 48 h,and the cells were treated with 2.5 mg/L DDP as positive control.The inhibitory rate was evaluated by MTT assay.Apoptotic rate was determined by TUNEL method.The expression of survivin and caspase-3 mRNA was detected by semi-quantitive RT-PCR.Results Compared with control group,the inhibitory rate was increased obviously(P<0.001),the apoptotic rate was increased markedly(P<0.001),the expression of caspase-3 mRNA was increased significantly(P<0.001),while survivin mRNA was decreased markedly(P<0.05 or P<0.01) in STE groups.Conclusion STE can induce MCF-7 cells apoptosis.The molecular mechanism might be related to up-regulating expression of caspase-3 and down-regulating expression of survivin genes.
