SD大鼠气管黏膜下注射硬化剂后VEGF表达及成纤维细胞的增生

目的:研究SD大鼠气管黏膜下层注射硬化剂对血管内皮生长因子(VEGF)表达及成纤维细胞增生的影响。方法:将54只SD大鼠随机分成正常组(6只)、对照组(24只)和硬化剂组(24只),后两组按第1、3、5和7天4个时间点各6只。正常组不作处理,对照组仅用空针刺激气管黏膜下层,硬化剂组用1%乙氧硬化醇0.05 ml气管黏膜下注射,分别在第1、3、5和7天取标本作VEGF免疫组化染色,用Image Pro Plus 5.1图像分析软件进行半定量分析积分光密度.并在电镜下观察成纤维细胞的增生变化。结果:(1)硬化剂组在4个时间点的VEGF积分光密度均明显高于对照组(P<0.01)。在硬化剂组内,第3天明显高于第1天,第3和第5天无明显差异,而第5天又高于第7天。(2)电镜下硬化剂组成纤维细胞从第3天开始明显增多,并有胶原纤维形成,第5天达到高峰,胶原纤维变致密,并出现弹力纤维,至第7天成纤维细胞开始破碎、分解。结论:硬化剂能够明显促进SD大鼠气管黏膜下层VEGF的表达和成纤维细胞的增生,作用高峰时间在第3～5天。

Expression of VEGF and Hyperplasia of Fibroblast in SD Rats'Trachea Submucosa after Injection with Polidocanol

Objective:To study the expression of vascular endothelial growth factor(VEGF)in SD Rats' trachea submucosa and to observe the fibroblast with microscope.Methods:54 SD Rats were randomly divided into normal group(n=6),empty needle injection group(n=24)and polidocanol injection group(n=24).The latter two groups were equally divided into four sub- groups according to the time of 1st day,3~(rd)day,5~(th)day and 7~(th)day.The Rats' trachea submucosa were injected with empty needle or 0.05ml 1% polidocanol,and the normal group without any interference.All the samples were stained by immunohis- toehemistry,and were observed by electron microscopes.The integrated optical density(IOD)of VEGF were analyzed with Im- age Pro Plus 5.1 software.Results:(1)The IOD of VEGF in the polidocanol injection group was higher than those in the emp- ty needle injection group(P<0.01).Within the polidoeanol injection group,the IOD of VEGF was much higher in the 3~(rd)day than in the 1~(st)day,and it was also higher in the 5~(th)day than in the 7~(th)day,but there was no difference between the 3rd and 5~(th) day.(2)According to the electron microscope,the fibroblast in the polidocanol injection group were much more than those in the normal group.Collagen fiber bundles present from the 3~(rd)day and became denser and more tightly packed from the 5~(th)day to the 7~(th)day.Conclusion:Polidocanol can distinctly increase the expression of VEGF in SD Rats' submucosa and visibly pro- mote the hyperplasia of fibroblast,and the apparently reaction time was in the 3~(rd)and 5~(th)day.
