高压氧对缺氧新生大鼠脑超微结构及细胞凋亡的影响

摘    要:目的观察高压氧治疗对新生大鼠缺氧损伤后脑超微结构及脑细胞凋亡的影响.方法新生30h的Wistar大鼠制作缺氧性脑损伤模型,随机分为缺氧后24h组、9d组和高压氧治疗组,并设正常对照组,采用流式细胞仪检测脑细胞凋亡率、电镜观察海马区超微结构.结果脑细胞凋亡率缺氧后24h和第9天分别为3.21%和1.32%,均明显高于正常对照组(0.16%)和经高压氧治疗9d组(0.57%).缺氧后24h海马神经细胞线粒体肿胀、脊断裂、高尔基体囊泡扩张,神经毡水肿明显;缺氧后9~18d,神经细胞固缩改变、双核仁,胞浆致密,线粒体肿胀,脑组织点状溶解灶,周围髓鞘变性,胶质细胞增生,微血管腔受压变形.经高压氧治疗9~18d后,海马脑组织除胶质细胞和突起轻微肿胀外,神经元和突触无明显异常.结论高压氧治疗可减轻缺氧所致脑损伤.

Abstract:Objective To observe the effects of the hyperbaric oxygen(HBO) on ultrastructure and apoptosis of neural cells of hippocampus in newborn rats with hypoxic ischemic encephalopathy (HIE).Methods Forty two wistar rats were divided into 4 groups: group 1 (for 24 hours after hypoxia),group 2 (for 9 days after hypoxia),group 3 ( treatment with hyperbaric oxygen) and group 4 (control group).The apoptosis of brain cell was detected with flow cytometer,the ultrastructure of hippocampus was detected with electron microscope.Results Apoptotic rates of brain cell in group 1 and group 2 were 3.21% and 1.32% respectively,both were significantly higher than these in group 3 (0.57%) and group 4(0.16%).In group 1,mitochondrion swelled and carina ruptured,Golgi apparatus vesicle expanded in hippocampus nerve cells and neuropil edema were observed;9 18 days post anoxia,neuronal shrinkage,nuclear membrane di nucleolus,mitochondria swelled,point lysis in brain constitution,chromoplasm denseness myelin sheath degeneration,glial cellular proliferation,microbloodvessel compression and deformation were observed.In treated group,neuron and synapse were normal except glial cell and enation of hippocampus constitution swelled lightly.Conclusion The brain injury for hypoxia may be relieved with HBO therapy.
