摘    要:目的 探讨趋化因子受体CCR7下调对95D肺癌细胞转移、侵袭的影响及相关机制。方法 95D肺癌细胞经不同剂量的白藜芦醇处理后,通过免疫荧光染色、PCR、Western印迹法等观察CCR7及其相关信号通路的表达。Transwell小室检测细胞侵袭和迁移能力。结果 白藜芦醇处理后95D细胞CCR7表达显著降低,与其相关的信号传导蛋白PI3K、p-AKT表达量亦降低,并呈剂量依赖性。Transwell小室结果证明经白藜芦醇处理后的肺癌细胞侵袭力下降,同时也抑制CCR7配体对肺癌细胞的趋化作用。结论 本研究结果提示白藜芦醇下调CCR7的表达同时也可下调PI3K、p-AKT表达,抑制肿瘤细胞侵袭转移的过程并促进其凋亡。

Abstract:Objective To investigate the effect of down-regulation of CCR7 expression on the invasion and migration ability in lung cancer 95D cells. Methods Human lung cancer 95D cells were treated with different does of resveratrol. The expression of CCR7 and related signaling pathway proteins were assayed by immunofluorescence, Real-Time PCR and Western blotting, respectively. The cell invasion and migration ability were evaluated by Transwell assay. ResultsThe expression of CCR7, PI3K, p-AKT protein in 95D cells were decreased in a dose-dependent manner after resveratrol treatment. Transwell array showed that invasion ability of 95D cells was inhibited with the decreasing of CCR7 expression, and cell migration induced by SLC also inhibited. Conclusion The results suggest that resveratrol can down-regulate expression of CCR7, PI3K and p-AKT, which might be involved in the inhibition of tumor cell invasion and metastasis, and enhance cell apoptosis.
