高压氧抑制外伤性角膜血管新生的实验研究

摘    要:目的 探讨高压氧对化学烧伤所致角膜血管新生(corneal neovascularization,CNV)的影响。方法 实验组与对照组各26只鼠(52眼),用硝酸银灼伤双眼角膜,实验组行高压氧治疗。伤后不同时期定量检测角膜中血管内皮生长因子(vascular endothelial growth,VEGF)的积分光密度(integration optic density,IOD),计数血管内皮细胞标记指数(labeling index,LI);伤后第7天用血管灌注、计算机图象分析测定CNV面积百分比。结果 第7天实验组CNV显著少于对照组(P<0.01);伤后两组角膜VEGF的IOD与血管皮细胞LI呈明显的平行关系,实验组各值均低于对照组对应值(P<0.01或P<0.05)。结论 高压氧治疗能降低角膜VEGF蛋白水平和血管细胞增殖水平,抑制CNV形成。

Abstract:Objective To study the effect of hyperbaric oxygen(HBO) on corneal neovascularization(CNV).Methods 52 Wistar rats were randomly didvided into two groups,26 rats (52 eyes) in each group.Both eyes were burned by silver intrate.The test group was treated with HBO.At different time after burn,internsity optic density(IOD) of vascular endothelial growth factor (VEF) protein in cornea was measured,and labeling index(LI) of vascular endotheliums was counted.At the 7th day,the vascular perfusing technique and computerized image analysis were used to determinate the ratio of the area of CNV to that of entire cornea.Results At the 7th day,CNV in test group was significantly less than that in control group (P<0.01).After burn,VEGF,IOD paralleled to LI of vascular endothelial cells in two groups,and the measurements in test group were less than that in control group at different day (P<0.01 or P<0.05).Conclusion HBO can effectively inhibit the level of VEGF protein in cornea after chemical burn and reduce the proliferation level of vascular endothelial cells,thereby inhibit the formation of CNV markedly.
