Abstract:Objective: The research on the quality evaluation of the aerial parts of Chuanxiong Rhizoma involves two aspects: fingerprint analysis and content determination. Methods: The fingerprint was performed on Kromasil C18(250 mm×4.6 mm, 5μm), the mobile phase was 0.1%phosphoric acid aqueous solution(A)-acetonitrile(B) at a rate of 1.0 mL/min, the sample size was 20μL, the column temperature was 35°C, and the detection wavelength was 254 nm. The content determination was performed on Kromasil C18(250 mm×4.6 mm, 5μm), the mobile phase was aqueous solution(A)-methanol(B)(34∶66) at a rate of 1.0 mL/min, the sample size was 10μL, the column temperature was 35°C, and the UV absorbance was monitored at 330 nm and 237 nm to determinate the content of ligustilide and n-butylidenephthalide. Results: There were 20 common peaks in the fingerprint of 13 batches of samples, with the similarities of more than 0.89. The content of ligustilide and n-butylidenephthalide of 13 batches of samples were 0.0642%~0.2633% and 0.0057%~0.0325%, respectively. Conclusion: The method is simple, accurate and reproducible, and can be used as a reference method for quality evaluation and quality control of the aerial parts of Chuanxiong Rhizoma.
