花椒不同炮制品水提液的传导麻醉作用研究

摘    要:目的比较花椒不同炮制品水提液对大鼠离体坐骨神经传导电位的影响,从而揭示花椒不同炮制品传导麻醉作用的差异。方法按照体重将Wistar大鼠随机分为空白组、溶剂组(水);花椒生品、清炒品、醋制品、盐制品、酒制品水回流提取液高、低2个剂量组(药物浓度0.2,0.1 g·mL-1);阳性对照组(利多卡因20 mg·mL-1),每组6只。剥离大鼠离体坐骨神经,用电刺激刺激远端神经干,利用PowerLab多通道生理检测仪记录正常动作电位与采用蘸有花椒不同炮制品(生品、清炒品、醋制品、酒制品、盐制品)水回流提取物的棉花包裹后的动作电位,观察空白组与给药组动作电位的振幅变化,以盐酸利多卡因为阳性对照分析数据。结果用蘸有花椒水回流提取物的棉花包裹后,大鼠离体坐骨神经动作电位的振幅产生显著变化,高、低电位差值如下:空白组(53.82±5.32)mV、溶剂组(52.97±6.34)mV、花椒生品低剂量组(25.42±7.51)mV、花椒生品高剂量组(16.80±6.68)mV、花椒清炒品低剂量组(32.56±8.71)mV、花椒清炒品高剂量组(29.69±7.72)mV、花椒醋制品低剂量组(41.59±6.92)mV、花椒醋制品高剂量组(31.40±7.42)mV、花椒盐制品低剂量组(24.95±7.19)mV、花椒盐制品高剂量组(13.36±5.54)mV、花椒酒制品低剂量组(41.20±6.21)mV、花椒酒制品高剂量组(32.49±6.25)mV和盐酸利多卡因组(9.06±3.86)mV。各药物组与空白组相比,高、低电位差均显著减小,各组局部麻醉作用由强到弱依次为盐酸利多卡因>花椒盐制品>花椒生品>花椒清炒品>花椒醋制品>花椒酒制品。结论花椒不同炮制品水提液均具有显著的局部麻醉作用,其中花椒盐制品传导麻醉作用最强。

Abstract:Objective To compare the effects of water extract from different processed products of Zanthoxylum bungeanum on the conduction potential of rat sciatica in vitro,so as to reveal the difference of conduction anaesthesia of different processed products of Zanthoxylon bungeanum.Methods According to weight,the Wistar rats were divided into blank group,solvent group(water),Zanthoxylum bungeanum raw group,stir-fried group,vinegar group,salt group,wine group,lidocaine group,high and low dosage group of Reflux extract from wine water(0.20,0.10 g·mL-1),positive control group(lidocaine 20 mg·mL-1),six rats in each group.Dissection of rat sciatic nerve in vitro.The isolated sciatic nerve of rats was stimulated by electrical stimulation.A Power Lab multi-channel physiological detector was used to record the action potential of cotton wrapped with different processed products(raw,fried,vinegar,wine,salt)dipped in pepper.The amplitude change of action potential in blank group and administration group were observed.Lidocaine hydrochloride was used for positive control analysis.Results In vitro sciatic nerve of rats,the amplitude of action potential of cotton wrapped with Zanthoxylum bungeanum water reflux extract changed significantly.The difference of high and low potential were as follows:Blank group(53.82±5.32)m V,solvent(water)(52.97±6.34)m V,raw low dose(25.42±7.51)m V,raw high dose(16.80±6.68)m V,stir-fried low dose(32.56±8.71)m V,stir-fried high dose(29.69±7.72)m V,vinegar low dose(41.59±6.92)m V,vinegar high dose(31.40±7.42)m V,salt low dose(24.95±7.19)m V,salt high dose(13.36±5.54)m V,wine low dose(41.20±6.21)m V,wine high dose(32.49±6.25)m V,lidocaine(9.06±3.86)m V.The high and low potential difference between each drug group and the blank group decreased,and the local anesthetic effect of each group was counted from strong to weak:lidocaine hydrochloride>Zanthoxylum bungeanum salt products>raw products>stir-fried products>vinegar products>wine products.Conclusion The water extract of different processed products of Zanthoxylum bungeanum has significant local anesthetic effect,among which Zanthoxylum bungeanum salt products have the strongest conduction anesthesia effect.
