胰岛素对高脂饮食小鼠皮下及附睾周围来源的脂肪细胞血浆纤溶酶原激活物抑制物-1、叉头框蛋白C2及叉头框蛋白O1表达的影响

摘    要:目的研究高脂饮食喂养小鼠附睾周围和皮下来源的脂肪细胞血浆纤溶酶原激活物抑制物-1(PAI-1)、叉头框蛋白C2(FOXC2)及叉头框蛋白O1(FOXO1)表达水平,探讨不同类型肥胖的机制。方法 20只小鼠随机分为对照组和高脂组,分别给予正常饮食和高脂饮食喂养12周。取其附睾周围及皮下脂肪组织,原代培养前脂肪细胞经分化后,Western blot法测定脂肪细胞中PAI-1的蛋白表达水平,并用RT-PCR法检测胰岛素作用前后高脂组附睾周围及皮下脂肪细胞FOXC2和FOXO1的mRNA表达水平。结果高脂组小鼠体重显著高于对照组(P〈0.05);组内比较小鼠附睾周围脂肪组织PAI-1表达显著高于皮下脂肪组织,差异有统计学意义(P〈0.05);胰岛素作用后,FOXC2 mRNA水平有所升高,附睾附近脂肪细胞升高明显,与皮下脂肪细胞相比,差异有统计学意义(P〈0.05);胰岛素作用后,FOXO1 mRNA表达有所降低,附睾附近脂肪细胞降低明显,与皮下脂肪细胞相比,差异统计学意义(P〈0.05)。结论高脂小鼠附睾周围和皮下来源的脂肪细胞的PAI-1表达不同,胰岛素作用后FOXC2及FOXO1的表达存在差别,这种差别可能是不同类型肥胖的机制之一。

Abstract:Objective To research the expression of PAI-1, FOXC2 and FOXO1 in adipocyte from subcutaneous and around the epididymis in mice feeding high-fat diet, in order to explore the mechanism of different types of obesity.Methods 20 mice were randomly divided into control group and high fat group. The control group was given normal diet and the high fat group was given high fat diet. After 12 weeks, rats epididymal adipose tissue was taken out to culture preadipocytes, and then differentiationed, the fat cell PAI-1 protein expression was detected by Western blot method. The FOXC2 and FOXO1 mRNA expression before and after insulin action in hyperlipidemia groupepididymal and subcutaneous fat cells around were detected by RT-PCR assay. Results Body weight in high fat group was significantly higher than the control group(P 〈0.05). Within the two groups, PAI-1 expression in adipocyte around the epididymis was significantly higher than the subcutaneous adipose tissue, the difference was statistically significant(P 〈0.05). After effects of insulin FOXC2 mRNA levels increased, epididymal fat cells increased obviously, there was statistically significant difference compared with subcutaneous fat cells(P 〈0.05); the expression of FOXO1 mRNA significantly decreased, epididymal fat cells decreased obviously, there was statisticaly significant difference compared with subcutaneous fat cells(P 〈0.05). Conclusion Different expression in PAI-1 from hyperlipidemic mice subcutaneous and visceral adipose cells, after the action of insulin, there are differences of FOXC2 and FOXO1 expressions. The difference may be one of mechanisms of different types of obesity.
