外源脱氧核苷酸对淋巴细胞DNA辐射损伤修复的影响

摘    要:目的探讨外源脱氧核苷酸对电离辐射损伤的人外周血淋巴细胞DNA修复的影响。方法用X射线照射人外周血淋巴细胞,诱导DNA损伤后分为2组培养:一组为对照组,另一组为脱氧核苷酸补充组(1和10 mmol/L)。结果核酸营养组与对照组相比,微核率和染色体畸变率无明显变化(P0.05),同时延长培养时间,微核率和染色体畸变率显著降低,但2组亦未表现出差异。结论体外单纯补充外源脱氧核苷酸并不能促进淋巴细胞DNA辐射损伤的修复。

Abstract:[Objective]To study the effect of exogenous deoxynucleotide on repair of radiation-induced DNA damage in peripheral blood lymphocytes.[Methods]The peripheral blood lymphocytes were treated with X-ray to induce DNA damage,and divided into two groups which included the control group and deoxynucleotide supplement group(1 mmol/L and 10 mmol/L).[Results]There was no significant difference in the micronucleus rate and chromosome aberration rate between the deoxynucleotide supplement group and the control group(P>0.05).The micronucleus rate and chromosome aberration rate obviously reduced with the increasing of incubation time,and the difference between two groups was not significant.[Conclusion]The simple supplement of exogenous deoxynucleotide can not promote the repair of radiation damage to DNA in lymphocyte.
