青春期糖尿病大鼠生殖腺内AR的基因表达和受体水平变化

摘    要:目的 观察青春期糖尿病大鼠生殖腺内雄激素受体的基因表达及蛋白水平变化。方法 用STZ诱导青春期大鼠糖尿病模型,实验分正常对照组、糖尿病组及胰岛素治疗组,分别以Northern blot及放射配基法检测睾丸、附睾、前列腺内雄激素受体的mRNA及蛋白水平。结果mRNA水平:生殖腺内各部位的雄激素受体mRNA表达三组间均无差异;蛋白水平:生殖腺内各部位糖尿病组的雄激素受体水平均显著低于正常组。结论 糖尿病在翻译水平干扰了雄激素受体的合成,使受体水平下降。

Abstract:Objective To investigate the changes of gene expression and protein level of androgen receptor in pubertal diabetic rat. Methods To induce the diabetic model rat using STZ, the rats were divided into three groups: control, diabetes and diabetes with insulin replacement group. Detect the mRNA and protein level of androgen receptor in testis,epididymis and prostate using Northern blot and radioactive method respectively. Results On the mRNA level: in all reproductive glands, there is no difference in mRNA expression of androgen receptor among the three groups;on the protein level:in all reproductive glands, the level of androgen receptor in diabetic group is significantly lower than that of control group. Conclusions The synthesize of androgen receptor is influenced on the translation level, which decreased the level of androgen receptor.
