肌苷对缺血再灌注脑组织VCAM-1表达的影响

摘    要:①目的 探讨肌苷对局灶性缺血再灌注后脑组织血管细胞间黏附分子-1(VCAM-1)表达的影响。②方法 将健康成年雌性SD大鼠70只随机分为假手术组、治疗组和对照组,治疗组在缺血再灌注前腹腔注射肌苷注射液,另两组腹腔注射相同体积的生理盐水。后两组分别在脑缺血90min后再灌注2h、6h、12h、1d、2d、3d、7d和14d等不同时间,用免疫组化技术观察VCAM-1的表达。③结果 在皮质区,VCAM-1的表达在缺血再灌注不同时间点呈现不同水平,假手术组和对照组缺血再灌注2h之前有痕量表达,VCAM-1的阳性表达在缺血再灌注6h有较明显增加,1~2d时表达达高峰,之后逐渐下降,到7d时VCAM-1的表达接近假手术组水平;在纹状体区呈现与皮质区相似的表达规律,但其表达与同时间点皮质区比较水平更高。在治疗组VCAM-1的表达规律类似于对照组,在同一时间点较对照组表达显著降低,又明显高于假手术组。④结论 适量输注肌苷可能通过VCAM-1作用机制对缺血再灌注脑组织起一定的保护作用。

Abstract:Objective To study the effects of inosine on the expression of vascular cell adhesion molecule 1(VCAM-1) in cerebra after focal cerebral ischemia and reperfusion. Methods Seventy adult female Sprague Dawley(SD) rats were randomly divided into sham-operation group (SG), and ischemia-reperfusion group(IG). Both SG and IG groups were subdivided into eight subgroups. The rats were made into middle cerebral artery occlusion (MCAO) model at 2 h, 6 h, 12 h, 1 d, 2 d, 3 d, 7 d and (14 d), respectively, after ischemia of 90 min. The expression of VCAM-1 was measured by immunohistochemistry (IHC). Results (In IG, the level) of ICAM-1 in cerebral cortex and striatum varied with the time of ischemia reperfusion. The trace expression of VCAM-1 occurred at 2 h, marked at 6 h, peak at 1-2 d and then decreased gradually, whereas they were hardly observed in SG. In TG, although the level of VCAM-1 also varied with the hours of ischemia-reperfusion, the number of cerebral positive cells was fewer than corresponded hours in IG and more notable than in SG. There was a significant difference between the IG and the TG. Conclusion Appropriate concentration of inosine might be beneficial to cerebral tissue after ischemia reperfusion.
