抑癌基因PTEN和血管内皮生长因子在大肠癌中的表达及相关性研究

目的探讨抑癌基因PTEN和血管内皮生长因子（VEGF）蛋白在大肠癌中表达的意义。方法采用免疫组化sP法检测48例大肠癌中PTEN和VEGF蛋白的表达，并分析其与临床和病理特点的关系。结果大肠癌中PTEN蛋白表达阳性率为45．8％（22／48），显著低于正常大肠黏膜组织的阳性率（P〈0．01）；VEGF蛋白表达阳性率为72．9％（35／48），显著高于正常大肠黏膜组织的阳性率（P〈0．01）；PTEN和VEGF蛋白表达呈负相关（P〈0．01）；PTEN蛋白低表达和VEGF蛋白高表达与大肠癌Dukes分期、浆膜浸润、淋巴结转移有关（均P〈0．01），而与年龄和组织学类型无关（P〉0．05）。结论PTEN和VEGF蛋白表达与大肠癌临床病理特征和生物学行为有密切关系，联合检测PTEN和VEGF蛋白的表达，对大肠癌的恶性程度及预后判断具有重要的临床意义。

Study on the significance of tumor suppressor gene PTEN and vasular endothelial growth factor (VEGF) protein expression in colorectal adenocarcinoma

Objective To investigate the significance of tumor suppressor gene PTEN and vasular endothelial growth factor （VEGF） protein expression in colorectal adenocarcinoma, Methods The expression of PTEN and VEGF protein was examined with immunohistochemical SP method. Results In colorectal adenocarcinoma, the positive expression rates of PTEN and VEGF proteins were 45.8% （ 22/48 ） and 72.9% （ 35/48 ）, showing the expressions of PTEN and VEGF proteins in cancer tissues were significant difference to theirs in normal mucosa （ P 〈0.01 ）. The expression of PTEN protein was negatively correlated with that of VEGF protein（ P 〈0.01 ） ,The low level of PTEN protein expression and the high level of VEGF protein expression were correlated with the Dukes stage, serosa infiltration, lymph node metastasis of colorectal adenocarcinoma（ P 〈 0.01 ）. They were not significantly correlated with patient/s age or histological type （ P 〉 0.01 ） Conclusion The exporessions of PTEN and VEGF protein wer significantly correlated with the clinicopathological characteristics and biologic hehaviors in colorectal adenocarcinoma. Combined detection of PTEN and VEGF may be helpful to evaluate malignant degree and prognosis of colorectal neoplasms.
