肝细胞癌有关肿瘤细胞调控基因差异表达的研究

摘    要:目的 探讨细胞周期调控与癌、抑癌基因在肝细胞癌 (hepatocellularcarcinoma ,HCC)中的表达。 方法提取 2 2例HCC患者肝癌及癌旁组织的mRNA ,逆转录合成含有3 3 p dATP标记的cDNA探针 ,与含有 3170个基因或EST(表达序列标签 )的cDNA微阵列进行杂交。结果 肝癌组织与癌旁组织间共有 136 9个差异表达基因或EST ,其中有 12 1个基因或EST表达差异 >2倍。肝癌组织与癌旁组织比较 ,12 1个基因或EST中表达上调的有88个 ,表达下调的有 33个 ;其中一些基因或EST在HCC中的表达上调尚属首次报道。结论 细胞周期调控与癌、抑癌基因参与了肝癌的发生 ,应用微阵列技术揭示了肝癌的基因表达谱 ,为研究肝癌的细胞周期调控与癌、抑癌基因提供了重要的线索。

Abstract:Objective To investigate cell cycle regulators, oncogenes and tumor suppressor genes expression in hepatocellular carcinoma (HCC). Methods The mRNA was extracted from cancerous and noncancerous liver tissues of 22 patients with HCC and synthesized into cDNA. The cDNA labeled with 33 P-dATP was hybridized for cDNA microarrays, each consisting of 3 170 genes or EST. Results The differential expression of 1 369 genes or EST was identified, including 121 genes or EST altered 2 times or more in cancerous tissues compared with noncancerous tissues. Compared with noncancerous tissues, 88 genes were overexpression and 33 genes were down-regulated, some genes was firstly reported in HCC. Conclusion The expression of cell cycle regulators, oncogenes and tumor suppressor genes are related to the mechanism of HCC, the information based on cDNA microarray analysis clarified genes expression profile and provide an important clue for cycle regulators, oncogenes and tumor suppressor genes.
