摘    要:目的:研究血管内皮生长因子反义寡核苷酸(VEGF antisense oligodeoxynucleotide,VEGF-ASODN)的聚乳酸聚乙醇酸(PLGA)纳米粒,并探讨其制备工艺.方法:采用复乳化溶剂挥干法制备纳米粒子,并用正交设计法对纳米粒的处方和制备工艺进行了优化.结果:纳米粒子形态圆整,大小均匀,平均粒径150 nm,包封率可达72%,含药量0.84%,体外释放缓慢,达到21 d.结论:VEGF-ASODN的PLGA纳米粒制备工艺简便、重现性好.

Abstract:Objective: To prepare VEGF antisense oligodeoxynucleotide(ASODN) loaded polylactic-co-glycolic acid(PLGA) nanoparticles and to study its physicochemical properties. Methods: PLGA nanoparticles were prepared with w/o/w double emulsion-solvent evaporation method by orthogonal design. Results: The shape of VEGF-ASODN loaded PLGA nanoparticles was round,the size was well-distributed,the mean diameter was 150 nm,the encapsulation efficiency reached 72 %,the VEGF-ASODN loading in the nanoparticles was 0.84% ,and the release rate was very slow,with the longest being 21 d. Conclusion: Our method for preparation of VEGF-ASODN loaded PLGA nanoparticles is simple and the reappearance is fine.
