叶绿素铜钠盐对AA模型基因表达谱的实验研究

摘    要:目的探讨叶绿素铜钠盐治疗再生障碍性贫血(AA)的作用机制。方法造成免疫介导 AA 小鼠模型,第15天从正常组、模型组及叶绿素铜钠盐组小鼠脾脏分别提取 mRNA,经反转录分别用 Cy_3、Cy_5荧光标记,获得 cDNA 探针,与博星基因芯片表达谱杂交,扫描仪扫描结果,用分析软件分析统计。结果模型组和正常组差异表达基因共100条(4.88%),其中 AA 时上调基因29条,下调基因71条。模型组和叶绿素铜钠盐组差异表达基因共53条(2.59%),其中上调基因25条,下调基因28条。结论叶绿素铜钠盐通过下调模型小鼠脾脏 H2-Bf 的 mRNA 基因表达,上调 CASH 基因表达达到治疗 AA 的目的。

Abstract:Objective To explore the effective mechanism of Chlorophyllin Capper Sodium Table(CCS)for AA.Methods Immune-in- duced AA mice model was established.Thirty Balb/c mice were randomly divided into 3 groups,therapeutic group was gasterogavaged by CCS, AA model group and normal group by salinn.Spleens were obtained from 3 groups on fifteenth day,the fluorescent labeled cDNA probes were pre- pared from mRNA which were extracted from spleens of normal group and AA group and CCS group.Two kinds of probes were then hybridized a- gainst Biostar Gene chip including 2048 genes,The fluorescent signals were scanned and analyzed.Results There were 100 differentially ex- pressed genes(4.88%)between normal group and AA group.The expression level of 71 genes was down-regulated while 29 genes up-regu- lated significantly in spleens in mice of immune-induced AA.There were 53 differentially expressed genes(2.59%)between CCS group and AA group.The expression level of 28 genes was down-regulated while 25 genes up-regulated significantly in spleens in mice of immune-in- duced AA.Conclusion CCS treats AA through decreasing the expression level of H2-Bf and increasing the expression level of CASH.
