羟基红花黄色素A对6-OHDA诱导的帕金森病小鼠模型的神经保护作用及机制研究

摘    要:目的 探讨羟基红花黄色素A对6-OHDA诱导的帕金森病小鼠模型的神经保护作用及机制。方法 通过旋转实验检测羟基红花黄色素A 对6-OHDA诱导的帕金森病小鼠模型的神经保护作用; 末次行为学试验后取脑,采用高效液相色谱-荧光检测法测定5-羟色胺(5-HT)水平; 采用试剂盒测定超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶( GSH-Px)、一氧化氮合酶(NOS)水平。结果 与空白组比较,对照组的旋转圈数明显增加(P<0.05),实验组的爬杆时间明显缩短(P<0.05); 使用羟基红花黄色素A治疗后实验组的旋转圈数明显降低(P<0.05); 羟基红花黄色素A能明显提高细胞中5HT、SOD、GSH-Px的水平,降低NOS 的水平,提高抗氧化能力。结论 羟基红花黄色素A对6-OHDA诱导的帕金森病小鼠模型具有神经保护作用,其机制可能与抗氧化作用有关

Abstract:ObjectiveTo study the neuroprotective effect and mechanism of hydroxysafflor yellow A on 6-OHDA-induced Parkinson mice model.Methods the neuroprotective effect of hydroxysafflor yellow A on 6-OHDA-induced Parkinson's mouse model was analyzed by rotation experiment. After the last behavioral experiment, the brains of mice were taken and the content of 5-hydroxytryptamine(5-HT)was determined by HPLC-fluorescence detection. The levels of superoxide dismutase(SOD), glutathione peroxidase(GSH PX)and nitric oxide were determined by kit. The changes of NOS level were analyzed and the mechanism of NOS was discussed.Results Compared with the blank group, the number of rotations in the control group was significantly increased(P< 0.05),the climbing time in the experimental group was significantly shortened(P< 0.05), after treatment with hydroxysafflor yellow A, the number of rotations in the experimental group was significantly reduced(P< 0.05). P-hydroxysafflor yellow A could significantly improve the levels of 5HT, SOD, GSH Px, reduce the level of NOS, and improve the antioxidant capacity.Conclusion p-hydroxysafflor yellow A had neuroprotective effect on 6-OHDA-induced Parkinson mice model, and its mechanism might be related to antioxidant effect.
