肝胃不和型功能性消化不良小鼠模型的建立

摘    要:目的:通过三联一复合应激法建立小鼠肝胃不和型功能性消化不良模型。方法:20只昆明小鼠适应性喂养一周后随机分为正常组和模型组,对模型组小鼠每天给予疲劳游泳、电击、慢性束缚和慢性不可预知温和应激刺激,连续干预30天。造模结束后用病理切片、胃残留率及相关脑肠肽水平评价胃肠功能,并结合糖水偏好试验、旷场试验、小鼠体重变化评估小鼠的精神状态。结果:与正常组相比,模型组的胃残留率、胃动素和p物质均显著下降,同时胃肠道无器质性病变,符合功能性消化不良的症状;小鼠体重、糖水消耗百分率和旷场试验得分均明显降低,说明小鼠精神状态不佳,符合肝胃不合的中医辨证。结论:通过复合应激法成功建立小鼠肝胃不和型功能性消化不良模型。

Establishment of a Novel Mouse Model of Functional Dyspepsia on Account of "Ganweibuhe" Syndrome

Abstract:Objective: To establish a mouse model with functional dyspepsia on account of " Ganweibuhe" syndrome by multiple stress method.Methods: After 1 week for mice to acclimatize the surroundings,20 Kunming mice were randomly divided into the control group and the model group. Mice in the model group received chronic unpredictable mild stress and three kinds of fixed stress: fatigue swimming、electric shock、and chronic restraint,for constantly 30 days. After modeling,pathological analysis、gastric residual rate and determination of relevant brain-gut peptide were carried out to evaluate the gastrointestinal function,while sugar preference assay,open-field test and body weight were performed to assess the mental state of mice. Results: Compared with the control group,gastric residual rate,motilin and substance P were significantly decreased in the model group( P < 0. 01) without any organic lesion in gastrointestinal tract,which was accorded with symptoms of functional dyspepsia. Meanwhile,the body weight,the percentage of sugar water consumption and scores of open-field test in the model group were lower than those in the control group,which revealed mice in the model group were in poor mental state,which was accorded with the diagnosis of liver-stomach disharmony in clinic of Traditional Chinese medicine. Conclusion: A novel mouse model of functional dyspepsia related to liver-stomach disharmony is successfully established by multiple stresses.
