姜黄素诱导人肝癌HepG2细胞发生凋亡及对细胞生长周期的影响

摘    要:目的研究中药姜黄素在体外诱导人肝癌HepG2细胞发生凋亡的机制及对细胞生长周期的影响。方法体外培养人肝癌HepG2细胞,用不同浓度的姜黄素(0.5、1.0、2.0、4.0、8.0和16.0μmol/L)刺激人肝癌HepG2细胞24、36和48h,MTT法及台盼蓝拒染法检测姜黄素对人肝癌HepG2细胞生长增值的影响并绘制生长曲线图,流式细胞术(FCM)检测其对细胞生长周期的影响。结果MTT法结果表明姜黄素对人肝癌细胞HepG2的生长具有抑制作用,2.0μmol/L浓度组在24h与对照组比较便具有统计学意义(P〈0.05),16μmol/L浓度组与HepG2细胞作用48h后对HepG2细胞的生长抑制作用最强(54.78±2.35)%,台盼蓝拒染法发现细胞存活率与姜黄素浓度间具有剂量和时间效应;流式细胞仪检测结果表明8.0μmol/L姜黄素诱导了人肝癌HepG2细胞发生凋亡,并对细胞生长的S期有明显的阻滞作用。结论姜黄素在体外能阻滞HepG2细胞的生长增值,诱导了人肝癌HepG2细胞发生凋亡。

Abstract:【Objective】To study the mechanism of Chinese herbal drugs curcumin on human hepatocarcinoma HepG2 cell and the influence to the vegetative cycle of HepG2. 【Methods】Cultured human hepatocarcinoma HepG2 cell in vitro and stimulated it with vary concentration of curcumin (0.5, 1.0, 2.0, 4.0, 8.0 and 16.0 μmol/L) for 24, 36 and 48 hours. MTT and trypan blue staining were carried out to detect the influence of curcumin to HepG2 cell multiplication and draw the cell growth curve, the cell vegetative cycle was detected by flow cytometic analysis.【Results】Vary concentration of curcumin has a obviously inhibitory action on the growth of HepG2 cell by MTT , there has a statistical significance with the concentration of 2.0μmol/L curcumin to the control group after 24 hours and the group of 16.0 μmol/L curcumin has a stronger inhibition (54.78±2.35)% to the HepG2 cell after 48 hours. There has a dose and time effect between the cell survival ratio and the concentration of curcumin by trypan blue staining. 8.0 μmol/L curcumin has induced HepG2 cell to apoptosis and has an apparent blocking effect on the S period of HepG2 by FCM. 【Conclusions】Curcumin can block the growth of human hepatocarcinoma HepG2 cell in vitro and has induced the cell to apoptosis.
