肝细胞生长因子对肺癌95D细胞系增殖和侵袭的影响

摘    要:目的探讨人类重组肝细胞生长因子(rhHGF)对人肺癌95D细胞系侵袭、增殖的影响及其机制。方法应用细胞培养技术培养95D细胞,分别用10,30,50μg/L不同浓度rhHGF作用95D细胞48h,以MTT法检测rhHGF对95D细胞增殖的影响;以过河实验、Transwell方法及RT-PCR方法检测rhHGF对95D细胞的运动侵袭能力和对MMP-9基因表达的影响。结果rhHGF可明显增加95D细胞的增殖和生长,且随浓度提高作用增强,呈剂量效应关系。过河实验、Transwell方法显示rhHGF能增强95D细胞的体外侵袭和运动能力,随浓度提高,过河时间缩短,细胞侵袭力增加(P〈0.05);RT-PCR测定显示rhHGF作用48h后,95D细胞中MMP-9基因表达上升。结论rhHGF可促进人肺癌95D细胞的生长和增加其体外侵袭能力,其机制与直接增加95D细胞迁移运动,及MMP-9表达增加有关。

Abstract:[ Objective ] To study the effect of recombinant human hepatocyte growth factor (rhHGF) on proliferation and invasion of 95D human lung cancer ceils and its underlying mechanism. [Methods] 95D cells were incubated with 10, 30, 50 μg/mL rhHGF for 48 hours. MTT assay was used to evaluate cell growth. The crossing-river test, Transwell method was used to examine the invasion of 95D cells. Expression of MMP-9 was determined by RT- PCR method. [ Results ] rhHGF promoted the growth and proliferation of 95D cells in a dose-dependent manner. The crossing-river time was shortened significantly and cell invasion increased (P 〈0.05). After the treatment with rhHGF for 48 hours, the expression of MMP-9 increased significantly (P 〈0.05). [ Conclusion] rhHGF promote the growth and increases the invasivc ability of lung cancer 95D cells. Its mechanism may be the increasing cell motility and up-regulating MMP-9 expression.
